肿瘤坏死因子相关凋亡诱导配体和顺铂对Hela细胞的诱导凋亡作用

摘　　要：目的观察不同浓度的肿瘤坏死因子相关凋亡诱导配体(TRAIL)和顺铂对Hela细胞的诱导凋亡作用。方法采用不同浓度的TRAIL作用于Hela细胞24 h,TRAIL100 ng/mL、顺铂10μmol/L及两者联合作用于细胞24 h,应用ELISA方法检测细胞凋亡率。结果不同浓度的TRAIL作用于细胞后,细胞凋亡率分别是10.91%、16.84%、22.79%、28.62%、34.85%,TRAIL、顺铂、及联合作用后Hela细胞的凋亡率分别为28.07%、46.13%、75.51%。结论同浓度的TRAIL对Hela细胞诱导凋亡发生具有明显的量-效关系,与顺铂联合应用可增加细胞对药物的敏感性。Objective To observe the effects of different dose of TRAIL and cisplatin on Hela cells.Methods Different concentration of TRAIL,TRAIL 100 ng/mL and cisplatin 10 μmol/L,combined TRAIL 100 ng/mL and cisplatin 10 μmol/L was treated on Hela cells for 24 hours respectively,the death rate of Hela cells was measured by ELISA.Results After being treated with different concentration TRAIL,Hela cells had death rates 10.91%,16.84%,22.79%,28.62%,34.85%;but after being treated with TRAIL,cisplatin,combined TRAIL and cisplatin,the Hela cells had death rates 28.07%,46.13%,75.51%.Conclusion TRAIL had obvious dose-effect on Hela cells and could increase the sensitivity of cells to drugs when combined with cisplatin.
