摘 要:目的探讨壮骨肾宝(淫羊藿、黄芪、白术)对维甲酸致骨质疏松大鼠不同部位骨骼的影响。方法 4月龄的SPF级雌性SD大鼠32只,随机分成正常对照组、维甲酸模型组、壮骨肾宝高剂量组、壮骨肾宝低剂量组,连续给药28 d。大鼠处死后取右侧股骨进行质量、骨密度的测量,取第4腰椎进行生物力学参数测量,取胫骨上段进行骨形态计量学测量。结果与正常对照组相比,维甲酸模型组大鼠股骨质量、骨密度均下降,腰椎生物力学参数弹性载荷、最大载荷、断裂载荷、刚度均下降,胫骨上段静态参数骨组织总面积(T.Ar)、骨小梁面积(Tb.Ar)、骨小梁周长(Tb.Pm)均降低,动态参数标记周长百分数、骨形成率均降低。与维甲酸模型组比较,壮骨肾宝低剂量可使弹性载荷增加,而高剂量使弹性载荷、最大载荷、断裂载荷、刚度均增加,但股骨质量、骨密度和胫骨上段骨形态计量学参数无明显变化。结论壮骨肾宝可预防维甲酸致大鼠腰椎生物力学性能的降低。AIM To investigate the effects of Zhuanggu Shenbao ( Epimedii Herba, Astragali Radix, Atractylo- dis macrocephalae Rhizoma) on different sites of the bones in rats induced by retinoic acid (RA). METHODS Thirty-two four-month-old virgin female Sprague-Dawley rats were randomly divided into control group, RA group, different doses of Zhuanggu Shenbao groups, being treated for 28 days. At the experimental endpoint, weight and bone mineral density (BMD) of femur, biomechanical properties of the forth lumbar vertebrae ( LV4), bone histo- morphometric parameters of the proximal tibial metaphysis (PTM) were performed. RESULTS Compared with the control group, weight and BMD of femur, biomechanical properties (elastic load, maximum load, break load, stifffness) significantly decreased in RA group. The static parameters (total tissue area, trabecular area, trabecular perimeter) and the dynamic parameters (% L. Pm, BFR/TV, BFR/BV and BFR/BS) significantly decreased in RA group. Compared with RA group, the elastic load of LV4 increased in low dose of Zhuanggu Shenbao group, biomechanical properties (elastic load, maximum load, break load, stiffness) of LV4 increased in high dose of Zhuanggu Shenbao group. Compared with RA group, there was no significant change in weight and BMD of femur, and histomorphometric parameters of PTM in all Zhuanggu Shenbao groups. CONCLUSION Zhuanggu Shenbao can increase the biomechanical properties of LV induced by RA in rats.
