摘  要:目的 :研究国产单硝酸异山梨醇酯 ( 5 ISMN)注射剂抗实验性心肌缺血及NO产生情况。方法 :观察药物对三种实验性心肌梗死的保护作用及多种离体血管条的效应 ,测定体内外给药其NO产生情况 (Griess法 )。结果 :5 ISMN能降低结扎大鼠LAD致心肌缺血的ΣST ,NST ,心肌缺血区百分比和血清CPK ,LDH(P <0 .0 5 ,0 .0 1) ;可使ISP诱发大鼠心肌缺血的ΣST及LVEDP降低 ,对血流动力学其他指标影响不明显 ;还可拮抗脑垂体后叶素所致豚鼠冠脉流量下降 (P <0 .0 5 ,0 .0 1)。本品对去甲肾上腺素 (NE)引起兔多种离体血管条的收缩呈浓度依赖性抑制作用 ,扩张静脉强于动脉。 5 ISMN体内外给药在无L Cys的磷酸缓冲液不能产生外 ,其他均有一定量NO产生 ,且呈剂量及时间依赖性 (P <0 .0 5 ,0 .0 1)。结论 :国产 5 ISMN注射剂具有一定抗心肌缺血效应及不能自发释放NO。OBJECTIVE:To study the effects of 5 ISMN on anti myocardial ischemia and nitric oxide release in animal models.METHODS:3 animal models of myocardial ischemia and many isolated vascular strips were adopted in the study and the release of nitric oxide in vitro and in vivo were determined by Griess method. RESULTS:5 ISMN may reduce the myocardial ischemic ΣST and NST,the percentage of ischemic area,and serum levels of CPK and LDH( P <0.05,0.01)induced by LDA ligation in rats;and it may also reduce the myocardial ischemic ΣST and LVEDP induced by ISP rats ( P <0.05,0.01) but no obvious effects on haemodynamic parameters.The drug may antagonize reduction in coronary blood volume( P <0.05,0.01)evoked by posterior pituitary in guinea pigs. The drug may inhibit the concentration dependent relaxation induced by NE in isolated strips of rabbits and this inhibitive effect was more pronounce in veins than in arteries .The generation of NO occurred in all cases when 5 ISMN is administrated,except in PBS or Krebs' solution without L cysteine. CONCLUSION:Domestic 5 ISMN injection has been shown to have significant anti myocardial ischemic effects but spontaneous release of NO do not occur.
