摘 要:目的:观察水芹乙酸乙酯提取物(简称水芹提取物)在乙型肝炎病毒(HBV)基因转染的人肝癌细胞系(HepG2)2215细胞培养中,对细胞的毒性及对HBV表面抗原(HBsAg)和e抗原(HBeAg)的抑制效果。方法:水芹提取物用培养液稀释成2000、1000、500、250、125μg·ml-1浓度,分别加入2215细胞内培养12d,观察药物对细胞的毒性。在无毒浓度下的水芹提取物,以培养液稀释1000、500、250μg·ml-13浓度,分别加入2215细胞内培养,于第6、9d收集的培养液,用固相放射免疫法测定HBsAg和HBeAg,γ-计数器测定每孔药液cpm值。结果:对细胞的半数中毒浓度(TC50)平均为2284.73±127.35μg·ml-1,最大无毒浓度为1000μg·ml-1。大、中剂量(1000、500μg·ml-1)对2215细胞分泌的HBsAg、HBeAg的活性于试验的第6、9d均有明显的抑制作用(P<0.05,P<0.01,P<0.001),并有一定转阴作用。结论:水芹提取物在2215细胞中培养6~9d,最大无毒浓度可明显抑制HBeAg和HBsAg的分泌。Aim To sduty the inhibitory effects of Ethyl Acetate Extract of Oenanthe Javanica on HbsAg and HbeAg and its toxicity in the cultured hepatocarcinomaline 2215 (Hep G 2) which was genetrasfered with hepatitis B virus.Methods Twenty-four hours after 2215 cells vaccinated,the different concentrations of Ethyl Acetate Extract of Oenanthe Javanica (2 000,1 000,500,250,125μg·ml -1 ) were added into culture fluid respectely, and exchanging the drugs every four days within 12 days.The cell alteration was observed under microcopy in order to evaluate the toxicity of Ethyl Acetate Extract of Oenanthe Javanica.The cells were treated with Ethyl Acetate Extract of Oenanthe Javanica (no toxicity concentration:1 000,500,250μg·ml -1 ) for 6 days and 9 days, culture fluid was collected and the contents of HBeAg and HBsAg were determined using radioimmunoassay kits respectively.Results Three seperated expirements showed that the mean half toxicity concentration of Ethyl Acetate Extract of Oenanthe Javanica was 2 284.73+127.35μg·ml -1 ) and its maximum of no toxicity concentration was 1 000μg·ml -1 ).Ethyl Acetate Extract of Oenanthe Javanica (maximum of no toxicity concentration:1 000,500μg·ml -1 ) significantly inhibited the secretion of HBeAg and HbsAg in cell line 2215 after 6 days and 9 days treatment(P<0.05 and P<0.01,P<0.001).Conclusion The results show that the maximum of no toxicity concentration of Ethyl Acetate Extract of Oenanthe Javanica can inhibite the secretion of HBeAg and HbsAg in cell line 2215 after 6 and 9 days.
