摘  要:为了检测不同浓度的HgCl2对泥鳅Misgurnus anguillicaudatus精子运动的影响,以泥鳅精子为实验材料,用含终浓度分别为0(对照)、1、5、10、15和20μmol/L的HgCl2待测液分别孵育0、2、4和6h后激活,激活后立即在显微镜(OlympusIX81)下观察精子运动参数。为进一步探讨HgCl2对泥鳅精子运动影响的机制,用终浓度为20μmol/L的HgCl2保存液孵育泥鳅精子10min,以含终浓度分别为0、0.1、1、10mmol/L的2-巯基乙醇和20μmol/LHgCl2混合液为激活液激活。激活后立即在显微镜下观察精子运动,发现2-巯基乙醇可逆转HgCl2对泥鳅精子的抑制作用,为探讨HgCl2对泥鳅精子运动影响的可能机制提供参考。The goal of this study is to investigate the motility characteristics of sperm in a gradient concentrations(0,1,5,10,15 and 20 μmol/L) of HgCl2 solution in loach Misgurnus anguillicaudatus.Firstly,loach was selected as the experimental material.Sperm were held in preserving fluid including HgCl2 at room temperature for 0,2,4 and 6 h,respectively,and sperm motility was observed by Olympus(IX81) after initiated.Secondly,to further observe 2-mercaptoethanol reversing the toxicity of HgCl2 to sperm motility,loach sperm were suspended in HgCl2 preserving fluid with the final concentration of 20 μmol/L.And the activating fluid was mixed with 2-mercaptoethanol and HgCl2.After mixed,the final concentrate of the HgCl2 was 20 μmol/L and the final concentrations of 2-mercaptoethanol were 0,0.1,1 and 10 mmol/L,respectively.After activation,2-mercaptoethanol is not only able to reverse the toxicity of HgCl2 to sperm motility,but also to enhance the effect(P〈0.01).Finally,the toxic mechanism of HgCl2 to loach sperm motility was discussed.
