摘  要:芦笋含糖颗粒剂中主要氨基酸为天门冬氨酸,采用薄层色谱扫描法测定其含量。影响测定的主要干扰是制剂中大量的糖。用商品磺酸型阳离子交换树脂预柱进行样品预处理,能较好地排除糖对氨基酸色谱的干扰,从而得到清晰的薄层图谱。用正丁醇-冰醋酸-水溶剂系统在纤维素板上展开,茚三酮试剂显色,在560nm作吸收扫描,天门冬氨酸在0.07~0.40μg范围内线性良好,回归方程:Y=40123.08X+3098.48,r=0.9989,平均加样回收率为97.8%,RSD=2.9%。原料芦笋浸膏及成品颗粒剂测定结果有良好的相关性,可作为植物药中氨基酸含量测定HPLC法以外的可供选择的检测方法。Aspartic acid is the main amino acid in Asparagus Granules. Determination of aspartic acid by means of TLC was reported in this paper. The sugar contained in the granules is the main factor that would seriously interfere the chromatogram. To clean up this interference, a commercial cation_exchange resin cartridege was used and the test solution was applied on a cellulose TLC plate, developed with the solvent system of butanol_acetic acid_water, and visualized with ninhydrin reagent. In situ TLC scanning in absorb mode under 560nm was conducted. The linear range is from 0.07 μg to 0.40 μg,the regression equation is Y= 40123.08 X+ 3098.49 (r= 0.99891 ), and the average recovery is 97.8% (RSD= 2.9 %).It is showed that the data obtained from the Granules and the raw material of Asparagus extract is closely interrelated. It is a reasonable alternative to determine amino acids by means of TLC other than HPLC method.
