摘 要:目的:观察知母提取物知母皂苷(TIM)及其苷元(SAR)对动物模型病理性升高的β肾上腺素受体的调整作用,同时观察SAR对家兔氢化可的松模型血清皮质醇含量以及正常小鼠和甲亢大鼠糖皮质激素受体密度的影响,旨在探讨两种提取物调整β肾上腺素受体的调整作用及其机制。方法:(1)观察TIM对甲亢大鼠模型脑组织β肾上腺素受体的调整作用;(2)观察SAR对家兔氢考模型外周淋巴细胞β受体的调整作用;(3)观察SAR对家兔氢考模型血清皮质醇含量的影响;(4)观察SAR对正常小鼠和甲亢大鼠糖皮质激素受体密度的影响;(5)用放射配基结合分析法检测动物组织细胞β受体和糖皮质激素受体数目(Rt值);(6)用放射免疫分析法测定血清皮质醇含量。结果:氢化可的松和甲状腺素可以分别使兔外周淋巴细胞和大鼠脑组织β受体数目升高;SAR和TIM可使上述动物病理性升高的β受体密度趋于正常;SAR不影响兔肝脏糖皮质激素受体密度和血清皮质醇(Cor)含量。结论:TIM和SAR对动物模型病理性升高的β受体的具有调整作用。OBJECTIVE:Regulative effects of sasapogenine (SAR) and Timosaponin (TIM) on number of binding sites of beta adrenoceptors of animal model were observed,to research the mechnism of these substances extracted from ZhiMu (a Chinses crud drug) regulating beta adrenoceptors, also investigated the influence of SAR on serum cortisol contents of rabbit hydrocortisol models and on glucocorticoid receptors (GcR) of normal mice and rat hyperthyroidism models.METHODS:(1) Observe the regulative effects of TIM on beta adrenoceptors of brain tissue in rat hyperthyroidism model.(2) Observe the regulative effects of SAR on beta adrenoceptors of lymphocytes in rabbit hydrocortisol models.(3) Investigate the influence of SAR on cortisol contents in serum of rabbit hydrocortisol models.(4) Investigate the influence of SAR on GcR of normal mice and rat hgperthyroidism models.(5) The density of beta adrenoceptors and GcR were measured with radioligand binding assay.(6) The cortisol contents in serum were detected with radioimmunine assay.RESULTS:Thyroxine and hydrocortisol could respectively manifold beta adrenoceptors of rabbit lymphocytes and rat brain tissue.SAR and TIM could correct elevatory numbers of binding sites of beta adrenoceptors of above animal models.SAR could not influence the binding sites of GcR and cortisol contents in serum of rabbit hydrocortisol models.CONCLUSIONS:TIM and SAR possess the regulative effects on beta adrenoceptors of animal models;that has nothing to do with cortisol contents and GcR.
