摘 要:目的:研究黄芪丹参有效组分不同比例配伍对肾纤维化模型大鼠肾小管上皮细胞转分化的影响。方法:将56只雄性SPF级大鼠随机分为正常组(n=8)、造模组(n=48)。造模组建立大鼠肾纤维化模型,分为模型组(n=8)、黄芪丹参药对组分1∶1配伍组(n=10)、黄芪丹参药对组分1∶2配伍组(n=10)、黄芪丹参药对组分2∶1配伍组(n=10)、福辛普利组(n=10)。造模后第2天开始各组给予相应干预措施,共干预14 d。观察大鼠肾组织病理变化,检测大鼠肾功能,并采用免疫组织化学方法检测大鼠肾组织上皮细胞钙黏蛋白(E-cadherin)、α-平滑肌肌动蛋白(α-SMA)的表达情况。结果:黄芪丹参有效组分不同剂量配伍各组对肾纤维化大鼠肾脏组织病理变化有不同程度的缓解作用。与正常组比较,模型组的Scr升高,差异有统计学意义(P<0.05),说明造模成功。与模型组比较,不同剂量配伍治疗组肾组织E-cadherin表达灰度值水平均明显降低(P<0.05),其阳性表达增强。而福辛普利组、组分配伍1∶1组肾组织α-SMA表达灰度值均明显升高(P<0.05),其阳性表达降低。结论:黄芪丹参药对有效组分1∶1配伍可以通过抑制肾小管上皮细胞转分化(EMT)而延缓肾纤维化的进展,其抑制转分化作用与福辛普利相当;其余不同剂量配伍对肾小管上皮细胞转分化抑制作用效果不明显。Objective: To investigate the effective components of Radix Astragali seu Hedysari and Radix Salviae Miltiorrhizae in different ratio on renal tubular epithelial cells transition in renal fibrosis rats models.Methods: Fifty-six female rats were randomly divided into seven groups: 8 rats in normal group,8 rats in model groups,10 rats in 1∶1 ratio group,10 rats in 1∶2 ratio group,10 rats in 2∶1 ratio group and 10 rats in Fosinpril group.Relevant intervening measures were carried out in each group for 14 days.The pathologic changes were observed,the renal function was detected,and the expressions of E-cadherin and α-SMA were detected by immunohistochemistry method.Results: Different ratio of Radix Astragali seu Hedysari and Radix Salviae Miltiorrhizae could relieve the pathologic changes differently in renal tissue of rats.The Cr in model group was higher than that in normal group,with a significant difference(P〈0.05).The expression level of grey value of E-cadherin in treatment group was decreased more significantly than that in model group(P〈0.05),and the positive expression was increased.The expression level of grey value of α-SMA in fosinpril group and 1∶1 group were increased obviously(P〈0.05),and the positive expression was decreased.Conclusion: Radix Astragali seu Hedysari and Radix Salviae Miltiorrhizae in 1∶1 ratio can retard the progression of renal fibrosis by restraining the transdifferentiation of renal tubular epithelial cells.The effects of 1∶1 ratio are equivalent to fosinpril.Inhibitory action of Radix Astragali seu Hedysari and Radix Salviae Miltiorrhizae in the other ratios have unconspicuous effects.
