酸性染料比色法测定飞龙掌血提取物中总生物碱的含量

摘　　要：目的:建立飞龙掌血提取物中总生物碱含量测定的方法。方法:通过酸性染料比色法,利用紫外分光光度计进行飞龙掌血总生物碱的含量测定,并进行方法学考察,建立适宜的含量测定方法。结果:建立了飞龙掌血总生物碱含量测定方法;于330 nm的检测波长下,以氯化两面针碱(Nitidine Chloride)为对照品,在1.497 6～5.990 4μg/ml范围内对照品含量与吸光度呈良好的线性关系;回归方程为A=0.115 9c+0.137 6(r=0.999 7);日内精密度RSD=0.53%、日间精密度RSD=2.57%;加样回收率为100.08%,RSD=3.30%。结论:该方法回收率、稳定性、重现性、精密度良好,可用于飞龙掌血总生物碱的含量测定。Objective： To establish a method for quantitative determination of the total alkaloids in Radix Toddaliae Asiaticae.Methods： Acid dye colorimetry and ultraviolet spectrophotometer were used to detect the content of total alkaloid in Radix Toddaliae Asiaticae with methodology research.The proper method for the determination was established.Results： The method for total alkaloid content determination was established;the result showed a good linear relationship between the absorbance and quantity of Nitidine Chloride to the total alkaloids by acid dye colorimetry at 330 nm wavelength.Within the range of（1.497 6~5.990 4）μg/ml,the regression equation was A=0.115 9c＋0.137 6（r=0.999 7）;the RSD of within-day precision was 0.53%,day to day precision was 2.57%;average recovery was 100.08%,RSD was 3.30%.Conclusion： This method is accurate,reliable and simple,which can be used for content determination of total alkaloids in Radix Toddaliae Asiaticae.
