血浆甘油三酯对酶联免疫吸附试验检测乙型肝炎病毒表面抗原的影响探讨

摘　　要：目的探讨甘油三酯(TG)对酶联免疫吸附试验(ELISA)检测乙型肝炎病毒表面抗原(HBsAg)的影响。方法收集HBsAg阳性血浆样本,用蒸馏水做对倍稀释至1∶8,分成4个组,对照组用蒸馏水稀释10倍,实验组用TG浓度不同的脂血浆稀释10倍。用ELISA法检测HBsAg的吸光度(A)值并做定性判读。结果试验组HBsAg的A值与对照组之间差异有统计学意义,试验组间HBsAg A值差异有统计学意义;HBsAg的定性判读试验组1与对照组间差异无统计学意义,试验组2、3与对照组间差异有统计学意义,试验组间差异有统计学意义。结论 TG浓度明显影响ELISA法检测HBsAg,TG浓度和A值有明显的相关性。在HBsAg滴度相同TG浓度不同的样本中,TG浓度增高,A值减低。在低滴度的HBsAg样本中,高TG浓度可导致A值低于临界值,出现假阴性。样本HBsAg滴度越低,高浓度TG的影响越明显。Objective To explore the effects of triglycerides on the enzyme-linked immunosorbent assay（ELISA） detection of HBsAg.Methods HBsAg-positive plasma samples,which were mixed with distilled water to 1∶8 times,were collected and divided into 4 groups;control group was diluted with distilled water to 10 times;the experimental groups were diluted with different triglycerides concentration of lipid plasma to 10 times.HBsAg absorbance（A） values were detected by ELISA and did qualitative interpretation.Results There were significant difference on HBsAg A values between test groups and control group.There were significant difference on HBsAg A values among different test groups.There were no significant difference on HBsAg qualitative interpretation between the test group 1 and control group.There were significant difference between the test group 2,3 and control group.There were significant differences among experimental groups.Conclusion The ELISA detection of HBsAg is significantly affected by triglycerides.There are significant correlation between triglycerides concentration and A values.A value is reduced on the basis of triglycerides concentration increasing in the same HBsAg titers.Triglycerides with high concentrations can lead to A values below the threshold in the low titer of HBsAg of sample,so as to show false negative.The lower of HBsAg titer,the more sereve on impact of triglycerides with high concentrations.
