益气化瘀解毒法对慢性萎缩性胃炎伴异型增生大鼠p53的影响

摘　　要：目的:观察益气化瘀解毒法对慢性萎缩性胃炎伴异型增生模型大鼠p53基因的影响.方法:Wistar大鼠60只随机分为造模组50只,空白组10只.采用以MNNG为主的三因素联合造模.28wk造模成功后,将造模组剩余的30只大鼠随机分为模型组、维酶素组、消痞颗粒组(10只/组).各组给予相应的药物.每天灌胃1次,连续治疗12wk后,Real-time PCR及Western blot法对各组大鼠p53基因进行检测.结果:消痞颗粒组野生型p53 mRNA表达量较模型组和维酶素组均显著增高(0.710±0.202vs0.443±0.081,0.456±0.148,均P<0.01);突变型P53蛋白表达量较模型组和维酶素组均显著降低(0.118±0.040vs0.886±0.127,0.350±0.065,均P<0.01).结论:中医益气化瘀解毒法可以逆转胃黏膜异型增生,这种作用可能是通过抑制p53基因的突变而实现的.AIM： To observe the effect of Yiqi Huayu Jiedu principle on p53 expression in chronic atrophic gastritis with dysplasia in rats. METHODS： Sixty Wistar rats were randomly divided into model group （n = 50） and blank group （n = 10）. Chronic atrophic gastritis with dysplasia was induced in rats of the model group. At week 28 after model induction, 30 rats in the control group were equally and randomly divided into model group, Weimeisu group and Xiaopi Granules group. Rats of each group were given the corresponding drug once daily. After treatment for 12 wk, the expression of p53 was detected byreal-time PCR and Western blot. RESULTS： The expression of wild-type p53 mRNA in the Xiaopi Granules group was signif- icantly higher than that in the control group and Weimeisu group （0.710 ± 0.202 vs 0.443 ± 0.081, 0.456 ± 0.148, both P 0.01）. The protein expres- sion of mutant p53 was signif icantly lower in the Xiaopi Granules group than in the control group and Weimeisu group （0.118 ± 0.040 vs 0.886 ± 0.127, 0.350 ± 0.065, both P 0.01）. CONCLUSION： Yiqi Huayu Jiedu principle can reverse gastric mucosal dysplasia possibly by inhibiting the mutation of p53 gene in rats with atrophic gastritis and dysplasia.
