甜蛋白thaumatin基因高效植物表达载体的构建

摘　　要：根据thaumatin氨基酸序列及植物蛋白质表达密码子的偏爱性,设计了thaumatinⅠ的全基因序列引物,采用重叠延伸PCR法,人工合成了thaumatinⅠ全长基因,克隆至载体pMD19-T上,序列测序表明,克隆的DNA序列与原设计序列完全相同,利用表达载体pRI101-ON,构建了高效植物表达载体pRI101-thaumatin。According to the amino acid sequence of the sweet protein gene thaumatin and plant biased codons,the whole gene sequence primer of thaumatin Ⅰ was designed.The whole gene of thaumatin Ⅰ was artificially synthesized by overlap extension PCR method,and then it was cloned to pMD19-T.The sequence analysis result showed that the synthesized DNA sequence was entirely accordant with the original designed sequence.The high-efficient plant expressive vector pRI101-thaumatin was finally constructed by using the expressive vector pRI101-ON.
