摘  要:目的:研究海螵蛸提取物对脲酶活性的抑制作用。方法:采用系统分离法即有机溶剂提取海螵蛸粉末,残渣再以热水浸提得到海螵蛸中的水提多糖,水提后残渣以氢氧化钠碱溶液获得碱提多糖,采用pH增值法首次探索了海螵蛸多糖等提取物对脲酶活性的体外抑制作用。结果:海螵蛸中多糖等有机物质对脲酶活性有一定的抑制作用。其中水提多糖和碱提水溶性多糖对脲酶活性的抑制作用较大,抑制率分别达到了70%以上,有机溶剂提取物中乙醚层和乙酸乙酯层提取物对脲酶活性的抑制作用在60%以上。结论:传统海洋中药海螵蛸中含有多糖等有机物质,以期对脲酶活性抑制作用的探索性研究为海螵蛸的临床应用机制和中成药开发提供一定的理论依据。Objective: To study the extract of cuttlebone and its inhibitory effect on Jack Bean Urease(JBU) activity.Method:The cuttlebone powder was firstly extracted by organic solvent,then the residue followed by hot water,and at last alkaline-solution extraction process was used to extract crude polysaccharides from the gived residue.The suppress activeness of the cuttlebone extract on JBU was explored by the pH value law.Result: Cuttlebone extract such as polysaccharides have some inhibitory effect on JBU.And the inhibitory effect of polysaccharides extracted respectively by water and sodium hydroxide solution are above 70%.The ethylether and ethyl acetate extract fractionated from the 95% ethanol soluble fraction also showed more 60% inhibitory effect on JBU.Conclusion: TCM cuttlebone contains organic compounds such as polysaccharides et al,and the study on the inhibitory effect of cuttlebone extract on JBU activity should be useful in clinical trials of pepticulcer and the development of Chinese Traditional Medicine.
