摘 要:分离培养Wistar新生大鼠的下丘脑神经细胞,在培养第7天以不同浓度的β-羟丁酸(β-hydroxybutyric acid,BHBA)(0、0.05、0.5、1、2mmol/L)处理24h,收集上清和细胞,提取细胞总RNA,分别利用荧光定量RT-PCR和ELISA的方法检测催乳素释放抑制因子(Prolactin release-inhibiting factor,PIF)基因表达和蛋白分泌的变化。结果显示,BHBA能显著抑制PIF基因表达和蛋白分泌,而且具有剂量依赖性。结果表明,BHBA可能做为信号分子调节下丘脑的分泌功能。β-hydroxybutyric acid(β-hydroxybutyric acid,BHBA)is not only a key precursor of synthesizing butterfat,but also an important signal molecule in vivo.In order to study whether BHBA can regulate the secretion function of hypothalamus,we isolate and culture hypothalamic neurons.At the seventh day of culturing,we treat cells with different concentration of BHBA(0,0.05,0.5,1 and 2 mmol/L).After 24 h,cells and culture supernatant were collected.The gene expression and secretion of PIF were detected with real-time PCR and ELISA methods.BHBA can significantly inhibit PIF gene expression and secretion in hypothalamic cells.The result suggested that BHBA may act as signal molecule regulating secretion function of hypothalamus.
