趋化因子CXCL12、CCL2、RANTES与早期流产的相关性

摘　　要：目的 探讨绒毛和蜕膜组织基质细胞衍生因子（ CXCL12）、单核细胞趋化蛋白-1（CCL2）、正常T细胞活化后表达和分泌的调节蛋白（RANTES）与早期流产的关系.方法选取天津医科大学第二医院2008至2009年收治的26例反复自然流产患者（自然流产组）、30例药物流产患者（药物流产组）和30例正常早孕妇女（对照组）绒毛和蜕膜组织,应用实时荧光RT-PCR方法检测3种趋化因子CXCL12、CCL2、RANTES-mRNA的表达.结果 早孕绒毛、蜕膜组织中均有趋化因子CXCL12、CCL2、RANTES-mRNA表达；CXCL12-mRNA的相对表达量自然流产和药物流产组绒毛（0.46±0.15,0.59±0.22）、蜕膜组织（0.35±0.13,0.42±0.17）均较对照组（1.79±0.82,0.81±0.21）显著降低（P＜0.01）,CCL2、RANTES-mRNA的表达均较对照组显著升高（P＜0.01）.结论 趋化因子CXCL12、CCL2、RANTES表达于早孕母胎界面绒毛和蜕膜组织中；绒毛、蜕膜组织CXCL12-mRNA低表达,CCL2、RANTES-mRNA高表达与早期流产的发生机制有关.Objective To explore the correlations between chemokines stromal cell-derived factor-1（CXCL12）,monocyte chemotactic protein-1 （CCL2）,regulated upon activation normal T cell expressed and secreted （RANTES） and early abortion. Methods The villus and decidua tissues of recurrent spontaneous abortion group （ n =26 ）,medical abortion group （ n =30） and normal early pregnancy women （ control group,n =30） were collected at our hospital from 2008 to 2009.And their expressions of CXCL12,CCL2,RANTES mRNA were determined by real-time fluorescence quantitative PCR （polymerase chain reaction）.Results The CXCL12,CCL2 and RANTES mRNA gene expressions were detected in firsttrimester placental villus and decidua tissues.And the gene expressions of CXCL12 mRNA in villus and decidua tissues in spontaneous abortion and medical abortion groups were significantly lower than that in the control group （P 〈 0.01 ）.But the gene expressions of CCL2 and RANTES mRNA in villus and decidua tissues were significantly higher than that in the control group （P 〈 0.01 ）.Conclusion CXCL12,CCL2 and RANTES are co-expressed in human villus and decidua tissues.The lower CXCL12 mRNA expression and the higher expressions of CCL2 and RANTES mRNA play important roles in the pathogenesis of early abortion.
