摘 要:目的探讨血必净对颅脑损伤的保护作用及机制。方法选取SD雄性大鼠108只,按随机数字表法分成假手术组、对照组、治疗组,每组36只,采用自由落体撞击伤方法制作创伤性脑损伤模型。治疗组大鼠伤后立即腹腔注射血必净4 mg/kg,每12小时重复1次。伤后24、72 h处死大鼠,用干、湿质量法测定脑含水量,测定MDA含量、SOD、GSH-Px活性及观察脑组织形态学改变。结果与假手术组比较,对照组、治疗组大鼠脑含水量、MDA含量均显著增高(P<0.01),SOD、GSH-Px活性显著下降(P<0.01);与对照组相比,治疗组大鼠脑含水量、MDA含量均显著下降(P<0.05),SOD活性显著增高(P<0.05)。结论血必净对大鼠创伤性脑损伤脑组织具有保护作用,其机制可能与减轻创伤后脑水肿、抑制氧自由基反应有关。Objective To investigate the protective effect of Xuebijing injection,an intravenous fluid derived from a traditional Chinese prescription,Xuefu Zhuyu Decoction,on traumatic brain injury(TBI) in rats.Methods A total of 108 Sprague-Dawley(SD) male rats were randomly divided into 3 groups(n=36 for each group),sham-operation group,injured group and the Xuebijing injection treatment group.The injured models of TBI were inflicted with free-falling objects(5.88 N·m).The animals from treatment group received an intraperitoneal injection of Xuebijing injection every 12 h(4 mg/kg) immediately after injury,and those out of the other 2 groups received the same route and dose of normal saline.Rats were sacrificed in 24 and 72 h after TBI.The water contents of brain tissue were measured with dry-wet weight method,and the activity of superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px),and the content of malondialdehyde(MDA) were detected with the biochemical techniques.Pathological changes of brain tissues were observed after HE staining.Results Compared with the sham-operation group,the levels of MDA were increased significantly(P0.01),the activities of SOD and GSH-Px were remarkably reduced(P0.01)in 24 and 72 h after TBI in the injured group and treatment group.But the above conditions in the treatment group were much better than in the injured group(P0.05).Conclusion Xuebijing injection protects rats against TBI,which might be due to its alleviating traumatic brain edema and inhibiting oxidative stress.
