氯化两面针碱固体脂质纳米粒大鼠体内药代动学研究

摘　　要：目的:比较氯化两面针碱固体脂质纳米粒和氯化两面针碱在大鼠体内的药动学。方法:12只Wistar大鼠随机分为2组,分别按剂量10mg/kg注射氯化两面针碱固体脂质纳米粒和氯化两面针碱,用高效液相法测定各时间点血药浓度,计算并分析药动学参数。结果:氯化两面针碱固体脂质纳米粒、氯化两面针碱T1/2分别为(276.9393±7.9606)、(163.519±9.9019)min;Cl(s)分别为(0.0599±0.0011)、(0.2037±0.0100)L.min/kg;AUC(0-t480)分别为(167.0805±3.2012)、(49.1851±2.3818)μg.min/ml。结论 :氯化两面针碱固体脂质纳米粒与氯化两面针碱相比,具有长效和缓释的特点。Objective：To compare the pharmacokinetics of nitidine chloride solid lipid nanoparticles（NC-SLN） and nitidine chloride（NC） in rats.Methods ：NC-SLN and NC were intravenously administrated to rats at dosage of 10mg/kg,the drug concentration were determined by the HPLC in different times,work out and analyze the pharmacokinetic parameters.Results ：The main pharmacokinetics paramters of NC-SLN and NC as follows：T1/2 were（276.9393±7.9606）,（163.519±9.9019） min resectively;Cl（s） were（0.0599±0.0011）,（0.2037±0.0100）L.min/kg,resectively;AUC（0-t480） were（167.0805±3.2012）,（49.1851±2.3818）μg.min/ml resectively.Conclusion：Comp are with NC,the NC-SLN contained characteristec of higher effect and sustained.
