摘  要:目的 :探讨补肾方、活血方、补肾活血方对血管性痴呆小鼠学习记忆能力及血浆血栓素B2(TXB2)和6-酮-前列腺素1α(6-Keto-PGF1α)含量的影响。方法:采用反复脑缺血再灌注法复制血管性痴呆小鼠模型,以尼莫地平为对照组,观察补肾方、活血方、补肾活血方三组小鼠水迷宫法行为学及血浆TXB2和6-Keto-PGF1α含量的变化。结果 :与假手术组比较,模型组小鼠学习与记忆能力明显下降(P<0.01),血浆TXB2含量明显升高,6-Keto-PGF1α含量明显下降(P<0.01);与模型组比较,各治疗组学习记忆成绩及血浆6-Keto-PGF1α含量均明显提高,血浆TXB2含量明显降低(P<0.05或P<0.01);各治疗组比较,补肾活血组(生药14.28g/kg)的学习和记忆成绩优于其它治疗组(P<0.05),补肾活血组和活血组(生药7.14g/kg)在降低血浆TXB2,升高6-Keto-PGF1α含量优于补肾组(生药7.14g/kg,P<0.05或P<0.01)。结论:补肾方、活血方、补肾活血方均可通过降低血浆TXB2,升高6-Keto-PGF1α含量,改善血管性痴呆小鼠学习与记忆能力,且补肾活血方与活血方在调节TXB2/6-Keto-PGF1α作用方面优于补肾方。Objective:To probe the effect of Bushenfang,Huoxuefang,and Bushenhuoxuefang on ability of learning and memory,and the content of TXB2 and 6-Keto-PGF1α in blood plasma of vascular dementia mouse.Methods:To reproduce the mouse model of vascular dementia through operation of cerebral ischemia reperfusion,and observe the praxiological changes by the means of water maze and the contents of TXB2 and 6-Keto-PGF1α in blood plasma of bushenfang,huoxuefang,and bushenhuoxuefang group mouse.Results:Compared with sham operation group,the ability of vascular dementia model mouse for learning and memory reduced,which showed extending the time for swimming the whole process,and increasing times of mistakes(P0.01),and the contents of TXB2 was decreased and 6-Keto-PGF1α was increased(P0.01).Compared with model group,the learning and memorizing performance record,and the contents of 6-Keto-PGF1α were improved,and the contents of TXB2 were decreased(P0.05 or P0.01)in each treatment group;the learning and memorizing performance record of the bushenhuoxue group(14.28g/kg crude drug)were excellent than other treatment group(P0.05),and the contents of TXB2 was decreased and 6-Keto-PGF1α was increased in the bushenhuoxue group and huoxue group(7.14g/kg crude drug) compared with bushen group(7.14g/kg crude drug,P0.05 or P0.01).Conclusion:The bushenfang,huoxuefang,and bushenhuoxuefang could decrease the contents of TXB2,and increase the contents of 6-Keto-PGF1α,so as to improve the learning and memory capability,and the regulation of TXB2 and 6-Keto-PGF1α in the bushenhuoxuefang,and the huoxuefang were excellent than bushenfang.
