摘 要:目的研究高胆固醇饮食对妊娠豚鼠胆囊运动及胆囊黏膜上皮水通道蛋白-1(AQP-1)和水通道蛋白-8(AQP-8)的影响。方法60只妊娠豚鼠,随机分为实验组A组、B组和对照组,实验组给予致结石饮食(胆固醇含量分别为1%和0.5%)喂养8周。采用RT-PCR对其胆囊AQP-1与AQP-8进行检测,应用化学发光法和酶联亲和抗体组织化学法分别检测血清雌二醇(E2)、孕酮(Pg)浓度及胆囊组织雌激素受体(ER)、孕激素受体(PR)的检测,同时测量各组豚鼠胆囊空腹体积(FV),胆囊空腹胆汁量(FB)的变化,并观察胆囊结石形成情况。结果A组、B组结石发生率分别为75%(15/20)、35%(7/20),明显高于对照组5%(1/20),且实验组结石均为以胆固醇类为主的结石。A组胆囊组织AQP-1与AQP-8mRNA表达低于B组,且显著低于对照组。A组其血清E2、Pg含量,胆囊组织ER、PR阳性表达率均高于B组,且明显高于对照组。同时A组FV及FB明显大于B组和对照组(P〈0.05)。结论高胆固醇饮食可降低妊娠豚鼠胆囊黏膜上皮AQPtmRNA与AQP8mRNA的表达,且可导致血清E2、Pg含量进一步升高,诱发胆囊运动功能下降,从而提高胆囊结石的发生率。Objective To explore the effects of high cholesterol diet(HCD) on the expressions of AQP- 1 mRNA and AQP- 8 mRNA in gallbladder and its role in the gallbladder motility in pregnant guinea pig. Methods Sixty pregnant guinea pigs were randomly divided into experimental group A and B and the control group. The experimental group A and B were fed with high cholesterol diet containing 1% cholesterol and 0.5% cholesterol respectively for 8 weeks. RT - PCR was used to evaluate AQP - 1 mRNA and AQP -8 mRNA in the gallbladder in all animals. The serum estrogen(E2) and progesterone(Pg) lev- el, and also estrogen and progesterone receptors in the gallbladder were evaluated by using chemilumines- cence and PAP immunohistochemical method in all animals. The gallbladder fasting volume(FV) and the gallbladder fasting bile volume (FB) were measured respectively. Results The incidences of gallstone in group A and B were 75% and 35%, which were significantly higher than the control one. The experimental animals all suffered with cholesterol gallstone. The mRNA expressions of AQP - 1 and AQP - 8 in gallblad- der of group A were lower than group B, and significantly lower than the control group. The levels of serum E2 and Pg, and also the positive expression rates of ER and PR in gallbladder between group A and B were significantly different. FV and FB in group A were significantly larger than group B and the control group (P 〈 O. 05). Conclusion HCD can decrease the mRNA levels of AQP - 1 and AQP - 8 in pregnant guinea pigg gallbladder and heighten the levels of serum E2 and Pg. It can also inhibit gallbladder motility obviously. This sequence of events in turn results in a significantly higher prevalence of cholesterol gallstones in the pregnant guinea pig fed by HCD.
