金贝乳康片对乳腺癌癌前病变肝郁肾虚模型大鼠的干预作用  

Intervention with Jinbei Rukang Tablet on the Liver Qi Stagnation and Ki dney Deficiency Model of Precancerous Breast Lesions

摘　　要：目的研究金贝乳康片对乳腺癌癌前病变肝郁肾虚模型大鼠的干预作用。方法将70只SD大鼠随机分为正常组(15只)、对照组(15只)、模型组(20只)、金贝乳康片组(20只)。用二甲基苯蒽(DMBA)联合慢性轻度不可预计应激及冷水刺激应激制备乳腺癌癌前病变肝郁肾虚模型。金贝乳康片组给予金贝乳康片水溶液灌胃1.5g/kg,其余各组予生理盐水。30天后腹腔取血并采集乳腺织织,HE染色观察病理学变化,ELLISA法检测血清皮质酮(CORT)、雌二醇(E2)及孕激素(P)水平。结果与正常组相比,模型组大鼠出现一系列生物表征变化,且CORT水平明显升高(P<0.05)。金贝乳康片组大鼠肉眼可见肿瘤发生率较模型组明显减少(P<0.05);病理以一般增生为主,较模型组明显减轻;血清CORT、E2水平较模型组明显降低(P<0.05),P水平略有升高(P<0.05)。结论金贝乳康片可降低肝郁肾虚证模型组大鼠肉眼可见肿瘤发生率,逆转乳腺癌癌前病变病理变化向一般增生转变,调节大鼠激素水平。Objective To explore the effect of Jinbei Rukang Tablet（Tablet for breast diseases） on the liver qi stagnation and kidney deficiency model of precancerous breast lesion.Methods The 70 SD rats were randomized into blank group（n=15）,control group（n=15）,model group（n=20）,and Jinbei Rukang Tablet group（n=20）.The liver qi stagnation and kidney deficiency model was prepared with DMBA jointed with multi-composite stress and cold w ater stress.Jinbei Rukang Tablet was given to the Jinbei Rukang Tablet group,while the other-groups were given normal saline.Thirty days later,bloo d collected from abdominal cavity and the breast tissue collected for pathologic al examination.HE staining was adopted for observing the pathological changes a nd ELLISA method adopted for detecting the level of corticosterone（CORT）,estra diol（E2）,and progesterone（P）.Results Comparing with the control group,there appeared a series of bi ological changes in the model group,and the level of CORT was significantly increased（P 0.05）.The incidence of visible tumors in the Jinbei Rukang Tablet group was s ignificantly less than that of the model group（P〈0.05）,the pathological c hange s characterized by proliferation were significantly relieved,and the levels of CORT and E2 were significantly decreased（P〈0.05） and the level of P was slightly increased（P〈0.05） than those of the model group.Conclusion Jinbei Rukang Tablet can reduce the incidence of visible tumors of t he rats of liver qi stagnation and kidney deficiency model group of precancerous breast lesions,promote the pathological changes of precancerous lesions changi ng to hyperplasia,and adjust hormone levels to inhibit progression of disease.
