翘芩清肺剂对哮喘大鼠平喘作用与变态及血管活性因子关系的研究

摘　　要：目的:研究翘芩清肺剂对哮喘模型大鼠的平喘作用及其与血小板活化因子(PAF)、白细胞总数(WBC)、嗜酸性粒细胞(EOS)及内皮素(ET)含量的变化关系,以探讨其作用机制。方法:将大鼠分为正常对照组、哮喘模型组、翘芩清肺剂高、中、低剂量组、地塞米松组和氨茶碱组,以卵白蛋白(OVA)建立哮喘大鼠模型。除正常组外,其余各组均在第1,8天以10%OVA 1 mL ip致敏,第15天起雾化吸入1%OVA激发哮喘,每天1次,每次20 min,连续2周,建立哮喘模型。从大鼠致敏第2天开始,各组大鼠以对应药物ig给药(其中地塞米松组从哮喘激发当日开始)。正常组、模型组给生理盐水5 mL.kg-1.d-1,地塞米松组0.27 mg.kg-1.d-1,氨茶碱组50 mg.kg-1.d-1,翘芩清肺剂高、中、低剂量组26,13,6.5 g.kg-1。造模时间为4周,取材后处死大鼠,观察药物对病鼠的哮喘程度、哮喘潜伏期的影响;测定血清PAF,WBC,EOS及肺泡灌洗液(BALF)中ET含量的变化。结果:翘芩清肺剂能有效减轻哮喘模型的哮喘程度(P<0.05)及明显延长病鼠的哮喘潜伏期(P<0.01),降低病鼠的PAF,WBC,EOS含量及ET水平,与模型组比较有显著性差异(P<0.05或P<0.01)。结论:翘芩清肺剂可有效改善哮喘状态,减轻哮喘发作,其作用机制可能与降低PAF,WBC,EOS及ET水平密切相关。Objective：To explore the mechanism of Qiaoqing Qingfeiji to relieving asthma of asthma model by studying the effect of Qiaoqin Qingfeiji on relieving asthma and the relation between it and the content of platelet-activating factor（PAF）,white blood cell（WBC）,eosinophils（EOS） and endothelin（ET） of asthma model.Method：Rats were divided into 6 groups：normal control group,asthmatic model group,Qiaoqin Qingfeiji with hight,meddle,and low dosage groups,and dexamethasone group,and aminophylline group.Asthmatic model was established by sensitizing and challenging rats with ovalbumin（OVA）.Except the normal control group,other groups were injected 10% OVA into abdominal cavities in the 1th day and the 8th day.2 weeks later followed by aerosolized 1% OVA once a day challenge to prepare the asthma models for 2 weeks.The drugs for every group were given intragastrically from the second day of rat sensitization for 4 weeks（the dexamethasone was given from the 1th day）.The effect of medicine on the asthma degree,asthma latency period and the content of PAF,WBC and EOS in serum,and ET in bronchoalveolar lavage fluid（BALF） of asthma model were observed.Result：Qiaoqin Qingfeiji can obviously improved the asthma degree,and extend asthma latency period（P0.05）,and obviously decrease the content of PAF,WBC and ET（P0.05-0.01）.Conclusion：Qiaoqin Qingfeiji can effectively recover from status asthmaticus,reduce the severity of asthma attacks.It is relate to reduce the content of PAF and WBC in serum,and ET in BALF of asthma model.
