摘　　要：目的探讨瑞舒伐他汀对动脉粥样硬化(AS)模型兔主动脉骨桥蛋白(OPN)的表达变化。方法复制AS兔模型,检测血清脂质成分;HE染色观察动脉的结构;用免疫组化和Western blot检测主动脉中OPN的表达。结果成功建立了AS兔模型;实验用兔在高脂喂养12周后血清中脂质相关指标有明显的增加(P<0.05);HE染色则显示了模型主动脉内膜增厚,有泡沫细胞生成并可见明显的AS斑块;免疫组化和Western blot结果表明主动脉平滑肌细胞的OPN表达显著上升(P<0.05),治疗后OPN表达降低,AS斑块减少。结论瑞舒伐他汀可能通过降低OPN的表达从而抑制AS的形成。Objective To study the effect of rosuvastatain on expression of osteopontin（OPN） in the artery wall of atherosclerotic（AS） rabbits.Methods The rabbit model of AS was induced by a high-cholesterol diet.We observed the structure of arteries by hematoxylin-eosin（HE） staining.OPN expression was detected by Western blot and immunohistochemical methods.Results The AS model was established successfully.The expression of OPN in the artery of high-lipid diet group increased markedly compared with that of control group.After treated with rosuvastatain in 12 weeks,the expression of OPN decreased compared with that of high-lipid diet group.Meanwhile,the HE staining indicated the foam cells products increased.Conclusion Rosuvastatain may decrease the expression of OPN,overexpression of OPN plays an important role in the AS.
