摘 要:[目的]优化亚临界水法催化制备微分子右旋糖酐的工艺。[方法]以右旋糖酐20为原料,采用亚临界水法催化制备微分子右旋糖酐,并在单因素试验的基础上,通过正交试验对右旋糖酐20水解的工艺条件进行优化。[结果]各因素对右旋糖酐20水解影响的大小顺序为温度>时间>固液比>搅拌转速。制备微分子右旋糖酐优选条件为反应温度170℃,反应1.5 h,固液比为0.6 g/ml,搅拌转速为200 r/min,制得微分子右旋糖酐重均分子量(Mw)为6 132,分布宽度(D)为1.67。[结论]该研究为制备微分子右旋糖酐提供了参考。[Objective] The aim was to optimize the micro-molecular dextran preparation based on sub-critical water catalytic.[Methods] Using sub-critical water catalysis to catalyze and prepare micro-molecular dextran from dextran 20.Furthermore,hydrolysis conditions of dextran 20 were optimized by orthogonal experimental design based on single factor experiment.[Result] Effects of each factor on the hydrolysis of dextran 20 were in order of temperature time solid-liquid ratio stirring speed.The optimum hydrolysis conditions of micro-molecular dextran were as follows: the temperature was 170 ℃;the reaction time was 1.5 h;the solid liquid ratio was 0.6 g/ml and the stirring speed was 200 r/min.Under the hydrolysis conditions,the molecular weight(Mw) of micro-molecular dextran was 6 132 and the dispersion of distribution was 1.67.[Conclusion] The research provides reference for the preparation of micro-molecular dextran.
