p27^(kipI)基因多态性与冠心病的相关性研究

摘　　要：目的:研究中国南方汉族人群细胞周期调控因子p27kipI基因多态性与冠心病的相关性。方法:采用基质辅助激光解吸电离飞行时间质谱技术(MALDI-TOF-MS)检测283例经冠状动脉造影确诊的冠心病患者和176例健康对照者的p27kipI基因-838C>A多态性,比较不同基因型和等位基因与冠心病患病风险的关系。结果:p27kipI-838C>A基因多态性等位基因和基因型的分布频率符合Hardy-Weinberg平衡(x2=2.472,P>0.05)。冠心病组-838A等位基因频率明显高于对照组(49.6%比40.5%,P<0.01)。Logistic回归分析发现,-838AA及-838AC基因型患冠心病的风险明显高于-838CC纯合子(P<0.01和P<0.05),且这一关系独立于性别、年龄、吸烟、高血压、糖尿病及血清TC、HDL-C、LDL-C水平等危险因素之外。结论:p27kipI-838C>A基因多态性可能是中国汉族人群冠心病发病的遗传危险因素之一。Objective：To investigate the correlation between the genetic polymorphism of p27kipI and susceptibility to coronary artery disease（CAD） in south Chinese Han population.Methods： Using matrix assisted laser desorption ionization time of flight mass spectrom-etry（MALDI-TOF-MS） methods,the genotypes and allels of p27kipI in 283 CAD patients diag-nosed by angiography and 176 healthy controls were analyzed.Results： The allel and genotype frequen-cies of p27kipI gene accorded with Hardy-Weinberg equilibrium（x2=2.472,P0.05）.the-838A allele in CAD patients was significantly higher than that of the control group（49.6% vs 40.5%,P0.01）.Compared with-838CC,-838AA and-838AC geno-types had a significantly increased CAD risk with the Logistic regression analysis（P0.01 and P0.05 respectively）.This correlation still existed after adjustment for the sex,age,smoke,hypertension,diabetes and plasma TG levels.Conclusion： The-838CA polymorphism in the p27kipI gene may contribute to an increased risk of CAD among south Chinese Han population.
