摘  要:目的:研究系统性红斑狼疮(SLE)MRL/lpr小鼠肾组织γδT淋巴细胞分泌细胞因子TGF-β1的功能。方法:流式细胞仪分析MRL/lpr小鼠外周血、肾组织γδT淋巴细胞比例,并体外选择性扩增γδT细胞,在体外培养条件下,测定培养体系中细胞因子TGF-β1的表达。结果:MRL/lpr小鼠外周血、肾组织中γδT淋巴细胞比例升高,肾组织中γδT淋巴细胞能异常分泌TGF-β1细胞因子。结论:肾组织γδT细胞可能通过分泌TGF-β1在狼疮性肾炎发病中起着免疫调节的保护性功能。Objective:To investigate the production of TGF-β1 by stimulated γδT cells in the kidneys from MRL/lpr mice with systemic lupus erythematosus.Methods:The proportion of γδT cells in peripheral blood and renal tissues in MRL/lpr mice were measured by flow cytometry.Then γδT cells was expanded in vitro to detemine the level of active TGF-β1 in supernatants by ELISA.Results:The proportion ofγδ T cells in peripheral blood mononuclear cells and the renal tissues from MRL/Ipr mice was significantly higher than those in the controls.The level of TGF-β1 produced by activated γδT cells in renal tissues from MRL/Ipr mice was significantly higher than that from the controls.Conclusions:γδT cells may play an important role in the pathogenesis of systemic lupus erythematosus via production of TGF-β1.
