疏风宣肺解毒汤抗甲1型流感病毒诱导细胞凋亡的研究

摘　　要：目的探讨疏风宣肺解毒汤对甲1型流感病毒感染的犬肾传代细胞凋亡的影响。方法将犬肾传代细胞分为六组:正常细胞对照组、病毒对照组、疏风宣肺解毒汤治疗组、疏风宣肺解毒汤对照组、奥司他韦治疗组、奥司他韦对照组,含药组均加入相应的最大无毒限量药液,Annexin V和PI双染用流式细胞仪(flow cytometry,FCM)测定细胞凋亡的百分率。结果疏风宣肺解毒汤治疗组和奥司他韦治疗组的凋亡率显著低于病毒对照组(P<0.01),而高于其药物对照组(P<0.01);疏风宣肺解毒汤对甲1流感病毒诱导的细胞凋亡的抑制率(IR)为67.07%,奥司他韦为76.36%。结论疏风宣肺解毒汤可抵抗甲1型流感病毒诱导的细胞凋亡,作用与奥司他韦相近。Objective To investigate the effects of Shufengxuanfeijieduedu Decoction on the apoptosis of Madin-Darby canine kidney（MDCK） cells infected by influenza virus A1.Methods MDCK cells was were divided into 6 groups：control group of normal cells,virus control group,Shufengxuanfeijieduedu Decoction treatment group,Shufengxuanfeijieduedu Decoction control group,Oseltamivir treatment group,Oseltamivir control group.Each group were added to the highest non-toxic doses of the corresponding liquide.The percentage of apoptotic MDCK cells was calculated through flow eytometry by Annexin V and PI co-dyeing.Results Shufengxuanfeijieduedu Decoction treatment group and Oseltamivir treatment group were significantly lower than the virus control group（P0.01）,but higher than their corresponding drug control group（P0.01）;the inhibition rate（IR） of influenza virus A1 induced MDCK cells apoptosis of Shufengxuanfeijieduedu Decoction was 67.07%,while the IR of Oseltamivir was 76.36%.Conclusion The results of the present study suggested that Shufengxuanfeijieduedu Decoction resist against the apoptosis induced by influenza virus A1 and have comparable effect to Oseltamivir.
