摘 要:目的探讨ADAM33基因多态性与吸烟人群COPD患者的相关性及其对COPD患者肺功能影响。方法利用PCR-RFLP方法检测COPD患者和健康对照者ADAM33基因中的4个位点单核苷酸多态性,分析ADAM33基因与COPD相关性及其对COPD肺功能影响。结果 T2G(P<0.001),T1G(P<0.001),S2C(P=0.03)等位基因的频率COPD组明显高于对照组;T1GG(P=0.0189)、T2GG(P=0.0026)基因型较野生型基因FEV1%预计值明显降低;T1GG(P<0.001)、T2GG(P<0.001)基因型TLCO%预计值较野生型明显下降。结论 ADAM33基因多态性与吸烟人群COPD有关,并对COPD患者肺功能产生了影响。Objective The aim of this study was to evaluate the relationship between polymorphisms of ADAM33 and smoker population with COPD,and it's influence to pulmonary function.Methods Four polymorphic loci(T2,T1,S2,S1) of ADAM33 were selected for genotyping.Genotypes were determined by using the polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) method.Results The distributions of the T2G,T1G,S2C alleles in COPD patients were statistically significant higher than controls(T2G P0.001;T1G P0.001;S2C P=0.03).Patients with T1GG and T2GG genotypes had much lower predicated value of FEV1% than wild genotype(p value:0.0189 and 0.0026,respectively);predicated value of TLCO% in T1GG and T2GG genotypes reduced significantly than wild genotype(p value0.001).Conclusions Polymorphisms of ADAM33 gene are associated with smoker population with COPD,and influence COPD patients'pulmonary function.
