摘  要:目的:观察细胞色素2J3(CYP2J3)基因转染对实验大鼠胸主动脉球囊损伤后狭窄程度的影响,分析上调CYP2J3/环-二十碳三烯酸(CYP2J3/EETs)系统拮抗血管损伤后狭窄的作用及其机制。方法:建立胸主动脉球囊损伤模型的同时分别经鼠尾静脉注入生理盐水、CYP2J3重组质粒和pcDNA3.1质粒(均为3ml/kg体重),分别为动脉损伤盐水组、动脉损伤CYP2J3组(动脉损伤2J3组)、动脉损伤pcDNA3.1组(动脉损伤3.1组);另取18只大鼠作为假手术组,不进行球囊损伤手术,只经鼠尾静脉注射与手术组相同量的生理盐水(假手术盐水组)、CYP2J3重组质粒(假手术2J3组)和pcDNA3.1质粒(假手术3.1组),每组均为6只大鼠。术后第28天取各组大鼠胸主动脉,分别检测动脉环相对管腔面积(RLA)、CYP2J3mRNA表达水平及11,12-EET含量。结果:动脉损伤各组RLA均小于对应假手术组(P<0.05或P<0.01),动脉损伤2J3组RLA大于动脉损伤盐水组及动脉损伤3.1组(P<0.01)。假手术2J3组CYP2J3mRNA表达高于假手术盐水组及假手术3.1组(P<0.05),动脉损伤各组CYP2J3mRNA表达及11,12-EET水平均高于对应假手术组(P<0.05或P<0.01),动脉损伤2J3组11,12-EET水平高于动脉损伤盐水组。结论:动脉损伤时CYP2J3/EETs系统上调可能是防止狭窄的重要机制。Objective:To investigate the impact of cytochrome 2J3(CYP2J3) gene transfection on experimented rat thoracic aorta stenosis after sac injury,and to analysis the effect and mechanism of up-regulating CYP2J3/epoxyeicosatrienoic acid(CYP2J3/EETs) system rivalried aortic stenosis after sac injury.Method:Establishing the thoracic aorta sac injury model,and meanwhile infusing physiologic saline,CYP2J3 recombinant plasmid and pcDNA3.1 plasmid through rat vena caudalis respectively(equally 3ml/kg),and so were injury physiologic saline group,injury CYP2J3 group and injury pcDNA3.1 group.Another taking 18 rats as sham-operated group,only infusing the same amount of physiologic saline(sham-operated saline group),CYP2J3 recombinant plasmid(sham-operated 2J3 group) and pcDNA3.1 plasmid(sham-operated 3.1 group) through rat vena caudalis,without sac injury.Each group were 6 rats.At the 28 days after aortic sac injury,the relative luminal area(RLA),CYP2J3 mRNA,11,12-EET levels of thoracic aorta were detected.Results:RLA of each injury group were less than the corresponding sham-operated group(P<0.05 or P<0.01),and RLA of injury 2J3 I group were higher than injury saline group and injury 3.1 group(P<0.01).CYP2J3 mRNA expression of sham-operated 2J3 group was higher than sham-operated saline group and sham-operated 3.1 group(P<0.05).CYP2J3 mRNA expression and 11,12-EET content of injury groups was higher than the corresponding sham-operated groups(P<0.05 or P<0.01),and 11,12-EET level of injury 2J3 group was higher than sham-operated 2J3 group and injury saline group.Conclusion:Up-regulating of CYP2J3/EETs system may be an important mechanism for prevention of thoracic aorta stenosis after sac injury.
