摘  要:探讨醋酸铅对脐带间质干细胞生物学特性的影响。用细胞计数法测定脐带间质干细胞活性,流式细胞仪检测醋酸铅对脐带间质干细胞凋亡的影响,用ALP组织染色分析成骨诱导的变化,并用RT-PCR和定量PCR分析醋酸铅作用前后的细胞因子表达情况。结果显示:脐带间质干细胞活力随醋酸铅浓度的升高而受抑,60μmol/L的醋酸铅可引起脐带间质干细胞凋亡率增加;成骨诱导分化后ALP表达率下降,TPO、SCF和VEGF等细胞因子表达也有所下降。因此,醋酸铅可以抑制脐带间质干细胞的增殖、促进其凋亡并影响其多向分化潜能。To explore the effect of lead acetate on the biological characteristics of hUC-MSCs,the effects of lead acetate on the activity of hUC-MSCs were investigated by cell count.Cell apoptosis was detected by FCM.We also studied the differentiation potency of MSCs through bone induction,and then detected the expression of ALP.TPO,SCF and VEGF were detected by RT-PCR or quantitative PCR.The results displayed that lead acetate can restrain the growth of MSCs.Lead acetate can increase the ratio of hUC-MSCs apoptosis at the concentration of 60 μmol/L.The ratio of ALP and cytokines expression was lower.This suggested that lead acetate can restrain the proliferation of hUC-MSCs,promote its apoptosis and degrade its multi-directional differentiation potentiality.
