摘  要:目的研究不同浓度醋酸铅染毒对大鼠脑细胞caspase家族表达的影响,探讨铅中毒诱导脑神经细胞凋亡的可能机制。方法40只Wistar大鼠随机分为4组:正常组(双蒸水,A组),高铅浓度组(2g/L,B组),中铅浓度组(1g/L,C组),低铅浓度组(0.5g/L,D组),各组大鼠分别自由饮用不同浓度的醋酸铅水溶液2个月;分光光度法检测脑组织匀浆液中caspase.3、caspase-8、caspase.9蛋白的含量。结果与A组大鼠比较,随染铅剂量增加B、C、D组大鼠脑组织中caspase-3、caspase-8、caspase-9蛋白含量显著增高(P〈0.001、P〈0.01、P〈0.05),并出现一定的浓度依赖性。结论铅中毒能明显增加caspase家族蛋白的表达,诱导大鼠脑神经细胞凋亡。Objective To research the effects of lead acetate with different concentration on caspase familial in cranial nerve cells of rats, and to investigate the possible mechanism of apoptosis in cranial nerve cells induced by lead poisoning. Method Fourty Wistar rats were divided into 4 groups randomly: normal group (distilled water, A group), high concentration group (2 g/L, B group), middle concentration group (1 g/L, C group), and low concentration group (0.5 g/L,C group), each group drunk lead acetate aqueous solution with different concentration for 2 months; the content of caspase-3, caspase-8, and caspase-9 proteins in brain tissue homogenate were detected by spectrophotometric method. Result Compared with A group, the content of caspase-3, caspase-8, caspase-9 protein in brain tissue of B, C, D groups increase significantly (P〈0.001, P〈0.01, P〈0.05) with concentration dependent. Conclusion Lead poisoning could increase the expression of caspase familial, and significantly induce apoptosis in cranial nerve cells.
