摘 要:目的:研究不同处理方法对人脱细胞脐动脉的组织结构影响,并比较其生物力学性质,为小口径组织工程人工血管支架的选择提供依据。方法:搏动条件下采用酶消化法脱去人脐动脉中的细胞成分(n=10),将每根脱细胞脐动脉平分为3段,随机均分为3组,A组经0.9%氯化钠液、B组经75%乙醇、C组经0.5%戊二醛处理。对处理后的脱细胞脐动脉进行HE染色、弹力纤维染色、生物力学检测(抗张力测试和抗爆破压测试),并进行比较。结果:脱细胞脐动脉经3种处理后B组及C组的塑形性较A组好;力学测试结果显示抗张力强度C组最大(4.63±1.06)N,B组次之(2.20±0.43)N,A组最小(0.93±0.33)N(P<0.05);抗爆破压强度B组(257±23)mmHg(1 mmHg=0.133 kPa),C组(261±23)mmHg无差异,均大于A组(143±19)mmHg(P<0.05);从位移-负荷曲线分析,C组的斜率最大,B组次之,A组最小;HE染色结果显示B组、C组脱细胞脐动脉的弹力纤维板形态能够很好地保存下来,而A组的弹力纤维板不明显;3组血管经抗张力测试,纵向拉伸或抗爆破压测试后,再采用弹力纤维染色进行形态学观察,A组的弹力纤维断裂比较明显,B组弹力纤维断裂较A组少,但较C组的断裂多。结论:使用75%乙醇或0.5%戊二醛处理后的脱细胞脐动脉塑性好,组织结构的形态佳、生物力学性质均明显较0.9%氯化钠液处理组好,而75%乙醇处理后的脱细胞脐动脉,在其柔韧性方面较0.5%戊二醛处理后更接近于生理状态,因此75%乙醇处理后的脱细胞脐动脉可能是较理想的血管支架材料。Objective:To compare the effects of different pretreatments on structures and biomechanical properties of human acellular umbilical artery,it could provide the basic selection of scaffolds in small diameter tissue engineering artificial blood vessel.Methods:Enzymatic digestion was used as a method to take off the cellular components of human umbilical artery(n=10) under the pulsatory conditions,each section of acellular umbilical artery was cut into equal thirds.Then they were randomly divided into three groups,and then group A was handled with 0.9% sodium chloride,group B was handled with 75% ethanol and group C was handled with 0.5% glutaraldehyde.Then all samples were studied by way of HE staining,elastic fiber staining,and bio-mechanical test including tensile and anti-burst pressure tests.Results: Biomechanical test results showed that tensile strength of group A,B and C were(0.93 ± 0.33) N,(2.20 ± 0.43) N and(4.63 ± 1.06) N respectively(P〈0.05);Burst pressure resistance strength of group A,B and C were(143 ± 19)mmHg(1mmHg=0.133 kPa),(257 ± 23) mmHg and(261 ± 23) mmHg respectively(P〈0.05).From displacement-load curve analysis,the slope of group C was the maximum,followed by group B,and group A.HE staining results showed that the elastic fiber morphology could be well preserved both in group B and C,but not in group A;From the observation of elastic fiber staining after tensile and anti-burst pressure tests,less broken elastic fibers were found in group B than group A,but more than group C.Conclusion: Pretreatment using 75% ethanol or 0.5% glutaraldehyde could retain better plasticity,morphological structure,and biomechanical properties in the acellular umbilical artery.The flexibility of 75% ethanol treated group was closer to the physiological state than 0.5% glutaraldehyde group.Therefore the acellular umbilical artery pretreated by 75% ethanol could possibly be a good choice as a scaffold in small diameter tissue engineering artificial blood vessel
