依达拉奉注射液Ⅰ型过敏反应试验

摘　　要：目的观察依达拉奉注射液经静脉注射给药后是否出现全身主动过敏反应和被动皮肤过敏反应。方法豚鼠分别腹腔注射依达拉奉注射液5mL(10mg).kg-1,0.9%NaCl注射液5mL.kg-1及牛血清白蛋白生理盐水注射液5mL(60mg).kg-1,隔日1次,连续3次进行致敏。末次致敏后12d各组静脉单次注射2倍剂量的以上相应受试物进行激发,观察30min内是否出现过敏反应;SD大鼠皮内注射相应抗血清0.1mL被动致敏48h后,分别静脉注射依达拉奉注射液5mL.kg-1,0.9%NaCl注射液5mL.kg-1和牛血清白蛋白生理盐水溶液10mL(100mg).kg-1,及1%伊文思蓝溶液1mL进行激发,30min后处死动物,观察注射点皮肤蓝斑直径。结果依达拉奉注射液致敏期间豚鼠未出现任何异常反应,给予激发剂量后亦未出现明显异常反应,未引起豚鼠死亡;依达拉奉注射液组和0.9%NaCl注射液组大鼠背部皮肤内层均未出现蓝斑。结论在该试验剂量条件下,依达拉奉注射液豚鼠全身过敏反应、大鼠被动皮肤过敏均为阴性。OBJECTIVE To observe edaravone injection after administration by inravenous injection of whether there is active systemic anaphylaxis and passive cutaneous anaphulaxis. METHODS Guinea pigs were injected edaravone injection 5mL（10mg） · kg^-1, 0.9 % NaCI injection 5mL·kg^-1 and bovine serum albuminsaline injection 5mL（60mg）·kg^-1, every other day, for 3 consecutive sensitization.Observe if there are allergic reaction within 30min, SD rats by intradermal injection of 0. lmL passively sensitized corresponding anti-serum after 48h, were intravenous edaravone injection 5mL· kg^-1, 0.9% NaCI injection 5mL·kg^-1 and bovine serum albumin 10mL（100mg）·kg^-1, and 1% Evans blue solution lmL to stimulate, 30rain after the death animals to observe the injection site skin and blue spot diameter. RESULTS Earavone injection of sensitized guinea pigs without any abnormality during the reaction, given dose failed to stimulate a distinct response, guinea pigs did not cause death, edaravone injection group and 0.9% NaCI injection in rats inner layer of skin back there were blue spots. CONCLUSION Under the conditions of the test dose, edaravone injection systemic allergic reaction in guinea pigs, rats were negative passive cutaneous allergy.
