摘 要:目的探讨柯萨奇B3病毒感染与HL-1心肌细胞凋亡的相关性。方法通过实时荧光PCR方法检测柯萨奇B3病毒感染4、12、24h及未感染的HL-1心肌细胞Bcl-2及Bax的表达,同时采用AnnexinV—PE凋亡检测试剂盒检测HL-1心肌细胞的凋亡率。结果柯萨奇B3病毒感染后4、12、24h细胞凋亡率均显著高于对照组(P〈0.01),感染后4h细胞凋亡率显著高于感染后12、24h(P〈0.01)。与对照组比较,柯萨奇B3病毒感染后4、24h的Bcl-2/Bax均明显变大(P〈O.05或0.01),而感染后12h的Bcl-2/Bax无明显变化(P〉0.05);感染后4h的Bcl-2/Bax显著大于感染后24h(P〈0.01)。结论柯萨奇B3病毒可导致HL-1心肌细胞的凋亡,在感染后4h其作用尤为明显。Objective To investigate the association of Coxsackievirus B3 infection with apoptosis of myocardial HL-1 cells. Methods The expression of Bcl-2 and Bax in HL-1 cells was detected by real time PCR, and apoptosis rate was examined by Annexin V-PE kit. Results Compared with non-infected HL-1 cells, the expression of Bcl-2/Bax in infected HL-1 cells increased significantly (P〈0.05)at 4h and 24h after infection; while Bcl-2/Bax expression in cells at 4h after infection was higher than that at 24h after infection (P〈0.05). Compared with non-infected HL-1 cells apoptosis rate in infected cells increased sig- nificantly(P〈0.05); while the apoptosis rate in HL-1 at 4h after infection significantly higher than those at 12h and 24h after infec- tion(P〈0.05 ). Conclusion Coxsackievirus B3 infection contributes to the apoptosis of HL-1 cells, especially 4h after infection.
