摘  要:采用抑制消减杂交方法结合targeted display法分离鉴定辛伐他汀处理高胆固醇血症小鼠后差异表达的基因,并采用定量RT-PCR方法对utrophin、TNF-α的mRNA转录水平进行了研究.结果表明辛伐他汀处理后7个基因上调表达,19个基因下调表达;这些基因涉及转录、脂质代谢、免疫反应等过程.定量RT-PCR分析显示utrophin与TNF-α的mRNA转录水平降低.Suppression subtractive hybridization (SSH) and targeted display (TD) combined to further identify and validate the differentially expressed genes. Seven genes after simvastatin treatment were upregulated and nineteen were down-regulated. The corresponding genes were found to be involved in transcription, lipid metabolism process, and immune reaction. The transcription of utrophin gene, coding for a cytoskeletal protein and sharing structural and functional homology with dystrophin, was found to be downregulated after simvastatin treatment. Utrophin mRNA levels in the liver were further analyzed using real-time quantitative RT-PCR. Tumor necrosis factor α (TNF-α) was simultaneously examined to get the information of inflammatory cytokines and utrophin. The results show that simvastatin decreases mRNA levels of utrophin and TNF-αin the livers of hypercholesteremic mice.
