摘 要:目的探讨聚焦超声二次辐照治疗小鼠宫颈癌U14移植瘤后对机体特异性抗肿瘤免疫功能的影响。方法 108只右侧腿部皮下成功接种的宫颈癌U14小鼠随机分为3组,每组36只。于接种肿瘤后第7天分别接受聚焦超声一次辐照治疗、聚焦超声二次辐照治疗及假照治疗;于接种肿瘤后第17 d,将各组分成2个亚组:分别在小鼠对侧(左侧)腿部皮下再次接种相同浓度的U14肿瘤细胞,记录各组小鼠的无癌存活数,以评价对同种肿瘤的抑瘤率(亚组1);用LDH法检测脾淋巴细胞毒性(亚组2)。结果与聚焦超声一次辐照组和假照组比较,聚焦超声二次辐照组对再次接种同种肿瘤的抑瘤率更高,体外脾淋巴细胞对同源肿瘤的杀伤活性更强,且随效靶比增加而增加。结论聚焦超声二次辐照治疗小鼠宫颈癌U14移植瘤后,相对于聚焦超声一次辐照治疗和假照治疗,机体发挥了更强的特异性抗肿瘤免疫作用。Objective To investigate the specific anti-tumor immune response after twice irradiation on cervical cancer U14 in mouse model. Methods 108 mice with cervical cancer cell U14 implanted in subcutaneous of right leg were divided into three groups, and 7 days after tumor implantation they receive one-time irradiation, twice irradiation and sham irradiation. 17 clays after tumor implantation, the mice in each group were divided into two sub-groups: re- inoculated the same concentration cancer cells U14 in subcutaneous of left leg, and then recorded tumor re-growth toevaluate the inhibition rate (group 1). Spleen lymphocytes were extracted to assess the cytotoxic activity against tumor cell U14 by using LDH assay(group 2). Results Compared with group of one-time irradiation and group of sham irra- diation, the group of twice irradiation had a higher rate of suppression for the tumor, and a significant difference of the cytotoxicity against U14 cells, the cytotoxicity enhanced while the ratio of the effort/target increased. Conclusions Compared with conventional one-time ablation, twice irradiation method could enhanced a stronger specific anti-tumor immunity.
