白藜芦醇甙对HIBD新生大鼠海马c-Jun表达的影响

摘  要:目的 探讨白藜芦醇甙对缺氧缺血性脑损伤(HIBD)新生大鼠大脑海马c Jun表达的影响.方法 选择新生7日龄SD大鼠52只,随机分为假手术对照组(NS组)、阴性对照组(NG组)、缺氧缺血组(HI组)、白藜芦醇甙干预组(PD组).通过结扎左颈总动脉及通入8%的氧气2小时制备新生大鼠缺氧缺血性脑损伤模型,应用免疫组织化学及蛋白质免疫印迹法测定缺氧缺血后24小时大鼠海马c Jun表达.结果 免疫组织化学结果 显示,HI组海马CA1、CA3区c Jun表达明显高于NS组和NG组(CA1区NS组t=15.39,NG组t=14.88;CA3区NS组t=11.16,NG组t=11.88,均P〈0.01),PD组海马CA1、CA3区c Jun表达明显低于HI组(CA1区t=12.61,CA3区t=10.22,均P〈0.01).蛋白质免疫印迹法结果 显示,HI组c Jun表达明显高于NS组和NG组(NS组t=33.67,NG组t=32.97,均P〈0.01),PD组c Jun表达明显低于HI组(t=29.78,P〈0.01).结论 白藜芦醇甙可能降低缺氧缺血性脑损伤新生大鼠大脑海马c Jun的表达,对于缺氧缺血后的神经元可能具有早期保护作用.Objective To investigate the effects of polydatin on expression of c-Jun in hippocampus of neonatal rats with hypoxic-ischenfia brain damage (HIBD). Methods Fifty-two 7-day old neonatal SD rats were randomly divided into 4 groups: normal sham-operated group (NS group), negative control group(NG group), HIBD group (HI group) and polydatin treatment group(PD group). The rat model of HIBD was established by ligating left carotid artery and providing 8% oxygen for 2hrs. hnmunohistochemistry and western blot were used to measure the expression of c-Jun in hippocampus 24 hrs after HI. Results The resuhs of immunohistochemistry demonstrated that the expression of c-Jun in hippocampal CA1 and CA3 region in HI group was significantly higher than that in NS group and NG group(t was 15.39 and 14.88, respectively, in CA1 region, and t was 11.16 and 11.88, respectively, in CA3 region, all P 〈0.01 ), and that the expression of c-Jun in hippocampal CAI and CA3 region in PD group was significantly lower than that in HI group( t was 12.61 in CA1 region and 10.22 in CA3 region, both P 〈 0.01 ). The results of western blot showed that the expression of c-Jun in HI group was signifieantly higher than that in NS group and NG group( t was 33.67 and 32.97, respectively, both P 〈 0.01 ) , and that the expression of c-Jun in PD group was signifieantly lower than that in HI group( t = 29.78, P 〈 0.01 ). Conclusion Polydatin can depress the expression of c-Jun in hippocampus of rats with HIBD, and might have protection effeet on HI neuron.
