摘 要:应用RNA干扰方法,设计合成了整合素的2个不同干扰位点的shRNA及空白对照载体,转化E.coli DH5α菌株,扩大培养后提取质粒转染猪脐静脉血管内皮细胞。RT-PCR方法检测显示,干扰载体在mRNA水平上的干扰效果明显。该干扰载体的构建及转染,为进一步研究整合素对猪细胞增殖特性及猪瘟病毒在细胞中的增殖作用奠定了基础。This research was designed to use RNA interference,currently more advanced methods,to design and synthetize the integrin's shRNA vector at two different sites as well as the control vector.Then we transfected them into E.coli DH5α strain in order to get the adequate amount of the plasmid before transfecting them into pig umbilical vein endothelial cells.We detected the expression of the integrin using RT-PCR and the electrophoretogram showed that the interference vector was successfully constructed and transfected into mammalian cells.The interference effect was obvious at the mRNA level.This also lay the foundation for further research of integrin's role in proliferation of the CSFV.
