摘 要:针对中药化学成分的"溶液结构"(即:溶液中溶剂化物质的微观结构及其宏观性质)与膜孔径的空间位阻,以黄连解毒汤等若干中医经典方及其多种指标成分为模型药物,通过原子力显微镜、激光动态光散射仪、高分辨核磁共振及其它分子生物物理学手段研究复方中药成分的"溶液结构"特征;借鉴超滤传质原理阐述膜过程中溶剂化效应所产生的中药成分竞争透过作用及机理;利用计算机化学技术探索中药体系"溶液结构"与膜材料微结构的相关性.融合多学科手段,深化基于整体观的中药复方现代基础研究,为构建可整体分离小分子药效物质组的中药专用膜分离技术、创新中药复方新药提供理论依据,解决中药复方制剂现代化的关键问题,推动中药学科与膜科学技术发展.Thousands years of experiences reveal that the water extract is the safest and effective carrier for traditional Chinese medicine. Membrane technology is a powerful tool for the cluster-screening of the small molecules from Chinese herbal. However, the lack of microscopic understanding of the water extracts of traditional Chinese medicine restricted the advantages of membrane technology. In this study, the key technologies to address these issues: the "solution structure" of traditional Chinese medicine chemical composinon (ie: the microstructure and macroscopic properties of the solvent material in solution) and the pore steric hindrance, a variety of index components of Huanglianjiedutang(HLJDT) as a model drug, by a) atomic force microscopy, dynamic laser light scattering, high-resolution nuclear magnetic resonance of molecular biophysics and other means of compound Chinese herbal medicinal ingredients of the "solution structure" characteristics; b) elaborating the mechanism of competitive filtration behavior by learning principles of solvent effects arising from the ultrafiltration membrane mass transfer process; c) exploring the correlation betwwen "'solution structure" of the chemical medicine system and the membrane material micro-structure by using the computer technology. The purpose of this study is: a) deepening overall concept-based modem research of Chinese herbal corr^ound by integrating of a multidisciplinary approach; b) establishing a specific membrane separation technology which can obtain small molecule groups from traditional Chinese medicine; c) providing a theoretical basis for new Chinese medidne; d) solving the key problems of Chinese herbal compound preparation and promoting the development of membrane science and technology.
