摘 要:[目的]观察游离脂肪酸(FFAs)对人成熟网膜脂肪细胞脂联素分泌的影响及罗格列酮(RSG)干预的效果.[方法]取6名外科手术女性患者的网膜脂肪组织,胶原酶消化分离纯化成熟脂肪细胞.细胞在FFAs(油酸∶软脂酸,2∶1)和(或)加用RSG的培基中培养不同的时间,酶联免疫吸附法测定培基中脂联素浓度.[结果]FFAs呈剂量及时间依赖性抑制网膜脂肪细胞脂联素分泌,各个干预组与对照组相比,脂联素降低有统计学意义;RSG可逆转此种抑制作用,高浓度(10 μmol/L)时逆转作用更强.[结论]FFAs能抑制体外培养的人网膜脂肪细胞脂联素分泌,但此种抑制作用可被RSG逆转.[Objective] To observe the effect of free fat acids(FFAs) on adiponectin secretion in human mature omental adipocytes and intervention by rosiglitazone(RSG). [Methods] Omental adipose tissue was ob- tained from 6 female patients undergoing elective surgery, and then digested, isolated and purified into mature adipocytes by collagenase treatment. Adipocytes were cultured in FEAs(oleate: palmitate, 2: 1) and/or RSG medium for different time. The adiponectin level in the medium was measured by ELISA. [Results] FFAs in- hibited adiponectin secretion in omental adipocytes in a dose- and time-dependent manner. Compared with con- trol group, the decrease of adiponectin in intervention groups had statistical significance. RSG could reverse the inhibitory effect. Moreover, high dose of RSG (10umol/L) had more prominent reverse effect. [Conclusion] FFAs can inhibit adiponectin secretion in cultured omental adipocytes. The inhibitory effect of FFAs can be reversed by RSG.
