摘 要:目的探讨皮质醇抑制血管新生的机制。方法将体外培养G0/G,期的人脐静脉内皮细胞(HUVEC)分为皮质醇组(无血清RPMll640培养液中加入皮质醇)和对照组(仅用无血清RPMll640培养液),细胞计数试剂盒(CCK-8)法检测细胞存活率,细胞迁移实验观察皮质醇对细胞迁移的影响,酶联免疫吸附法测定细胞中人碱性成纤维细胞生长因子(bFGF)和基质细胞衍生因子1(SDFl)含量,蛋白质印迹法测定细胞缺氧诱导因子1仅(HIF-1d)的表达。结果皮质醇组细胞活力与对照组相比无明显差异;细胞迁移数目明显少于对照组,差异有统计学意义[(618±117)个比(12004-57)个,P=0.00011];bFGF、SDFl与NO水平明均明显低于对照组[bFGF为(69.74-5.7)ng/L比(99.3±11.0)ng/L,P〈0.01;SDFl为(10.2±2.1)ng/L比(12.4±2.5)ng/L,P〈0.05;NO为(58.5±10.3)tzmol/L比(97.3±6.2)Ixmol/L,P=0.03407]。结论皮质醇对HUVEC细胞活力无明显抑制作用,但可以明显抑制其迁移功能,并降低细胞中NO、bFGF、SDFl的含量,这可能是皮质醇抑制HUVEC迁移的作用机制。Objective To investigate the effects of cortisol on the migration of human umbilical vein endothelial cells (HUVEC). Methods The viability of the cells was determined by using cell counting kit-8 ( CCK-8 ). Transwell_boydom system was used to test the migration of human umbilical vein endothelial cell induced by cortisol. Enzyme-linked immunosorbent assay(ELISA) method was used to determine the effect of cortisol on the expression ofBasic fibroblast growth factor(bFGF) and stromal cell derived factor-1 (SDF1). The expression of HIFI(x was analyzed with Western blotting. Results It showed that cortisol itself had no significant inhibition on cell viability of HUVEC but significantly inhibited HUVEC migration and decreased the expression of bFGF and SDF1. There were no differences of expression of Hypoxia-inducible factor(HIFlct) between control and cortisol groups. Conclusion Cortisol can decrease the expression of bFGF and SDF1 in HUVEC and inhibit the migration of HUVEC.
