摘  要:目的探讨制何首乌含药血清对造血祖细胞CFU-GM增殖的影响及制首乌对贫血大鼠骨髓细胞黏附分子表达的影响。方法制备制何首乌大鼠含药血清,体外半固体培养骨髓CFU-GM细胞;建立大鼠血虚动物模型,用流式细胞仪检测给药后大鼠骨髓单个核细胞CD44、CD54的表达水平。结果含药血清组大鼠骨髓CFU-GM集落数明显增多,与空白血清组比较差异显著(P<0.05);给药组CD44的表达与模型组无明显差异(P>0.05),但给药组CD54的表达显著低于模型组(P<0.05),且接近正常。结论制何首乌能促进造血祖细胞CFU-GM的增殖,并可改善贫血大鼠骨髓细胞黏附分于CD54的表达水平。Objective To study the proliferation efficiency of Polygoni Multiflori Preparata induced by medicated - serum on hematopoietic progenitor cell of colony- forming unit- granulocyte macrophage( CFU -GM) and the expressions of bone marrow cellular adhesion molecules in anemia rats treated with Polygoni Muhiflori Preparata. Methods We investigate the approach of cuhi- vating the CFU - GM of rat by semisolid in vitro, establish the model of blood deficiency from rats. Meanwhile, determine the expressions of CD44 and CD54 in bone marrow mononuclear cells detected by flow cytometry after administration. Results The colony numbers of CFU - GM treated with medicated - serum were obviously increased( P 〈 0.05 ) compared with the control. No signifi- cant difference between the administration group and the model control group for CD44 expression(P 〉 0.05 ), but for CD54 expression,there was significant difference between them(P 〈 0.05)and we found that the CD54 expression of administration group, which almost get to a normal level, were lower than those of the model control group. Conclusion Not only the proliferation of CFU - GM can be promoted, but also the CD54 expression levels can be up - regulated by Polygoni Multiflori Preparata.
