经前平颗粒对PMS肝气逆证模型大鼠顶区和额区皮质中μ阿片受体表达的影响

摘　　要：目的探讨PMS肝气逆证与μ阿片受体表达量的关系以及经前平颗粒的微观作用机制。方法以脉冲电刺激法制备PMS肝气逆证大鼠模型,运用半定量RT-PCR和Western blot技术检测PMS肝气逆证模型大鼠顶区和额区皮质中μ阿片受体的表达量。结果 PMS肝气逆证模型大鼠顶区和额区皮质中μ阿片受体的表达量均显著降低,给予经前平颗粒后能显著提高该基因表达。结论 PMS肝气逆证的发生可能与脑中枢顶区和额区皮质中μ阿片受体表达量下降有关,调节该基因表达异常可能是经前平颗粒的部分药理作用靶点。Objective By observing the expression level of μ-opioid receptor（MOR） in parietal and frontal cortex of PMS model rats with Liver-＂qi＂ invasion,this study investigated the relationship between PMS model rats with Liver-＂qi＂ invasion and MOR expression,and the intervention mechanism of Jingqianping granule. Methods We analyzed the expression of MOR in parietal and frontal cortex of PMS model rats with Liver-＂qi＂ invasion by Western Blot and semiquantitative RT-PCR. Results The expression of MOR in parietal and frontal cortex of model rats decreased obviously compared with the normal control group（P0.01,P0.05）.Compared with the model group,the expression of MOR increased after treated with Jingqianping granule（P0.01,P0.05）. Conclusion The mRNA and protein expression level of MOR in parietal and frontal cortex of PMS model rats with liver-＂qi＂ invasion were significantly lower than normal control rat,but when we gave the Chinese medicine Jingqianping granule the expression level of MOR in parietal and frontal cortex were notably higher than model group.So we infer that the expression changes of MOR are related to restlessness and tantrum of premenarche,also related to aggressive behaviour of premenarche.The Jingqianping granule can up-regulate the MOR expression level of the model rats in parietal and frontal cortex,which may be one of the mechanism to treat PMS with liver-＂qi＂ invasion.
