摘 要:目的:应用PCR-RFLP方法对天津地区96名汉族人进行HLA-DQA1等位基因的分型研究,以了解天津地区汉族人群HLA-DQA1多态性的分布。方法:通过PCR反应扩增HLA-DOA1第二外显子多态性片段,以限制性内切酶Dde Ⅰ、Hae Ⅲ、Rsa Ⅰ、Fok Ⅰ及Mnl ;Ⅰ、Mbo Ⅱ进行酶切反应,对HLA-DQA1基因进行分型。结果:在天津地区汉族人群中共检出8种DOA1等位基因,其中HLA-DQA1*0301出现频率最高(28.13%),DQA1*0401出现频率最低(0.52%)。结论:通过与其他人群HLA-DQA1分布特点进行比较得出,DQA1*0301和*0401是中国汉族人群HLA-DQA1基因中2个最具特点的等位基因。Purpose:To understand the polymorphism of HLA-DQAl in Tianjin population,HLA-DQAl alleles were determined in 96 unrelated Tianjin Han individuals using polymerase chain reaction-restriction fragment length polymorphism (PCR-RELP) method. Methods:PCR amplification of the polymorphic segment of HLA-DQAl second exon was performed ac-cording to the 11th International HLA Workshop Protocols. Amplified DNAs were digested with DdeⅠ,Hae Ⅲ,Fok Ⅰ,Rsa Ⅰ,MnI Ⅰ and Mbo Ⅱ. Results:Eight different alleles were detected in Tianjin Han individuals. Of the 8 DQAl alleles tested, the gene frequency of DQAl 0 301 (28.13%) was the highest, and that of DQAl * 0 401 (0.52%) was the rarest. Con-clusion: By comparing with other population, it was found that DQAl * 0 301 and * 0 401 were the characteristic alleles among the HLA-DQAl alleles in Han people.
