摘  要:目的 评价霉酚酸酯 (MMF)与雷帕霉素 (RAPA)合用预防及逆转大鼠心脏移植后排斥反应的作用。方法 将实验动物随机分为 4组 :(1)对照组 ;(2 )单用MMF组 ;(3)单用RAPA组 ;(4)MMF与RAPA合用组。结果 单用MMF和单用RAPA组与对照组比较均显著延长移植心脏存活时间 ,MMF与RAPA合用组与对照组比较非常显著地延长移植心脏存活时间 ,并且心脏存活时间明显长于同剂量两药的单用组。结论 MMF与RAPA具有很强的协同效应 ,能够预防与逆转大鼠心脏移植排斥反应。Objective To evaluate the role of combined use of mycophenolate mofetil (MMF) and rapamycin (RAPA) in prevention of acute heart allograft rejection and reversal of ongoing cardiac allograft rejection in rat.Methods The rats were randomly divided into control group and MMF group, RAPA group and MMF+RAPA group. For prevention of the acute rejection, the drugs were given from day 1 to day 30 by gavage; for reversal of the ongoing acute rejection, the drugs were given from day 4 to day 34 by gavage.Results For prevention of the acute rejection and reversal of the ongoing acute rejection, subtherapeutic doses of MMF and PAPA significantly prolonged cardiac allograft survival as compared with control group. Both drugs had a synergism interaction.Conclusion Combination of MMF and RAPA with subtherapeutic doses could produce a synergistic effect in prevention of acute cardiac allograft rejection and reversal of ongoing acute cardiac allograft rejection in the rat.
