乙酰肝素酶的RNA干扰对B16黑素瘤细胞实验性肝转移的影响

摘  要:目的研究用RNA干扰的方法抑制乙酰肝素酶的表达及其对B16黑色素瘤细胞实验性肝脏转移的影响。方法将HPA siRNA转入B16黑色素瘤细胞,抑制HPA的表达,并测定该细胞的增殖能力及黑色素生成能力;还将RNA干扰的B16黑色素瘤细胞注入小鼠脾脏测定其实验性肝转移的能力,并与对照组比较。结果将HPA siRNA转入B16黑色素瘤细胞,行RNA干扰后获得HPA较低表达的细胞(表达水平为对照组的20%,P<0.01),而黑色素生成及细胞增殖能力不受影响(P>0.05)。将该细胞注入小鼠脾脏后,计数实验性肝转移结节的数目,实验组较对照组肝脏转移结节明显减少(P<0.01)。结论 B16黑色素瘤细胞经RNA干扰后,随着HPA表达的降低,细胞转移能力下降。RNA干扰抑制HPA的表达,是一种理想的抗肿瘤治疗方法。[Objective] To study the RNA interference method in suppressing heparanase mRNA expression level, and the effect on the experimental hepatic metastasis caused by B16 melanoma ceils. [Methods] Transfect the HPA siRNA into B16 nelanoma cells to supress the HPA expression, and to evaluate the melanogenic and cell proliferate ability of transfected cells. [Results] Transfected with HPA siRNA, the HPA mRNA expression level in B16 melanoma cells was suppressed to 20% compared to that of control ceils (P〈 0.01), while the melanogenic and cell proliferate ability of transfected cells were comparable to control cells (P 〉0.05). When the HPA suppressed cells were intraspleniely injected into mice, much less experimental hepatical metastasis nodules were found compared to that of control (P〈0.01). [Conclusion] Treated with RNA interference, the metastatic ability of B16 melanoma cells decreased, that is well correlated with the suppressed HPA mRNA expression. RNA interference method to suppress HPA expression can be used as an anti-tumor therapitic strategy.
