摘 要:目的观察大鼠双侧基底外侧杏仁核(BLA)脑区微量注射去甲肾上腺素(NE)对丙泊酚遗忘效应的影响。方法将50只健康成年雄性Sprague-Dawley大鼠随机分为5组,每组10只。S+S组,双侧BLA注射0.9%氯化钠溶液+腹腔注射0.9%氯化钠溶液;S+P组,双侧BLA注射0.9%氯化钠溶液+腹腔注射25mg/kg丙泊酚;NE0.3+P组,双侧BLA注射NE0.3μg+腹腔注射25mg/kg丙泊酚;NE1.0+P组,双侧BLA注射NE1.0μg+腹腔注射25mg/kg丙泊酚;NE3.0+P组,双侧BLA注射NE3.0μg+腹腔注射25mg/kg丙泊酚。每组大鼠于连续被动回避实验训练前5min腹腔注射0.9%氯化钠溶液或25mg/kg的丙泊酚,于训练后即刻双侧BLA微量注射0.9%氯化钠溶液或不同剂量的NE,于训练后24h检测潜伏期及开场实验中的水平穿格次数及站立次数。结果与S+S组相比,S+P组、NE0.3+P组被动回避实验训练后24h的潜伏期显著缩短(P值均<0.05),而NE1.0+P组、NE3.0+P组的差异均无统计学意义(P值均>0.05)。各组大鼠的水平穿格次数及站立次数的差异均无统计学意义(P值均>0.05)。结论 BLA去甲肾上腺素能系统参与了静脉麻醉药物丙泊酚的遗忘效应。Objective To investigate the influence of norepinephrine(NE) micro-injection in bilateral basolateral amygdala(BLA) on the propofol-induced amnesia in rats.Methods Fifty male adult Sprague-Dawley rats were randomly divided into 5 groups(n=10).S+S group received normal saline injection both in BLA and intraperitoneally;S+P group received normal saline injection in BLA and 25 mg/kg propofol intraperitoneally;NE0.3+P group received 0.3 μg NE injection in BLA and 25 mg/kg propofol intraperitoneally;NE1.0+P group received 1.0 μg NE injection in BLA and 25 mg/kg propofol intraperitoneally;and NE3.0+P group received 3.0 μg NE injection in BLA and 25 mg/kg propofol intraperitoneally.Either normal saline or propofol(25 mg/kg) was intraperitoneally injected 5 min before continuous multiple-training of inhibition avoidance(CMIA),and normal saline or NE(0.3,1.0,or 3.0 μg) was injected into bilateral BLA immediately after training.Twenty-four hours later,the retention latency and the number of crossings and rearings in Open Field test were measured.Results Compared with S+S group,the retention latencies of S+P group and NE0.3+P group were significantly shortened(both P0.05),and the retention latencies of NE1.0+P group and NE3.0+P group had no significant difference(both P0.05).There were no significant differences in the number of crossings and rearings among the 5 groups(all P0.05).Conclusion The noradrenergic system in the BLA is involved in the propofol-induced amnesia.
