摘 要:目的:建立高效液相色谱法(HPLC)测定浙产虎杖根茎与根中大黄素含量的方法,并比较根茎与根中的大黄素含量。方法:采用超声提取法制备供试品溶液,运用高效液相色谱法测定虎杖根茎与根中大黄素含量。Zorbax XDB-C18色谱柱;流动相为甲醇-1%冰醋酸(70∶30);检测波长:254nm;流速:1.5 mL.min-1;柱温:40℃;进样量:20μL。结果:大黄素进样在1μg.mL-1~500μg.mL-1浓度范围内与峰面积线性关系良好(r=0.9995),加样回收率为97.56%,RSD为2.72%(n=6)。浙产虎杖根茎与根中大黄素含量分别为22.965mg.g-1和16.285mg.g-1。结论:本法适用于虎杖中大黄素含量的检测。浙产虎杖根茎与根中大黄素含量丰富且存在差异。Objective:To determine and compare emodin in rhizomes and roots of Polygonum cuspidatum Sieb. et Zucc. from Zhejiang province. Methods:The sample solutions were prepared by Ultrasonic extraction. HPLC method was used to determine emodin in rhizomes and roots of Polygonum cuspidatum Sieb. et Zucc. Zorbax XDB - C18 column was used and the mobile phase was CH3 -1% CH3COOH (70 : 30). The detection wavelength was 254nm, the flow rate was 1.5 mL · min^-1, the column temperature was 40℃, and the injection volume was 20μL. Results: Emodin showed a good linear(r = 0. 9995 )relationship at the range of 1 g · mL-1- 500g· mL-1, and its average recovery rate was 97.56% ( RSD = 2.72%, n = 6). The contents of emodin in rhizomes and roots of Polygonum cuspidatum Sieb. et Zucc. from Zhejiang province were 22. 965mg · g^-1 and 16. 285mg · g^-1. Conclusion:The method is suitable for the determination of emodin in Polygonum cuspidatum Sieb. et Zucc. The contents of emodin both rich in rhizomes and roots of Polygonum cuspidatum Sieb. et Zucc. from Zhejiang province and the contents of emodin in rhizomes and roots are different.
