枸杞籽油对C57BL/6J小鼠2型糖尿病肾脏损伤干预作用的研究

摘　　要：目的研究枸杞籽油对C57BL/6小鼠2型糖尿病肾脏损伤的干预作用。方法采用以高脂饲料饲喂后的C57BL/6小鼠,单次腹腔注射链脲佐菌素70mg.10g-1,建立小鼠2型糖尿病模型。设正常对照组、糖尿病肾损伤模型组、二甲双胍(3.0mg.10g-1)治疗组、枸杞籽油(0.05、0.1 mL.10g-1)两个剂量治疗组。给药5周后检测各组小鼠给药后的血糖、尿糖2、4h尿量2、4h尿蛋白定量、肾质量指数、肾小球面积、肾脏白介素-8含量、肾脏病理变化。结果枸杞籽油各剂量组与糖尿病模型组比较,可降低2型糖尿病模型小鼠的血糖、尿糖、24h尿量和尿蛋白定量、肾质量指数、肾小球面积、肾脏白介素-8含量,肾脏病变得到明显改变(P<0.05或0.01)。结论枸杞籽油能降低C57BL/6小鼠2型糖尿病的血糖、尿糖、24h尿量和尿蛋白定量、肾质量指数、肾小球面积、肾组织白介素-8的含量,具有防治糖尿病肾损伤发生的作用。Objective To study the renal lesion projective effect of Lycium Barbarum seed oil in C57BL/6J mice with type 2 Diabetic.Methods The animal model of type 2 diadetes renal lesion was established by once intraperitoneal injection of streptozocin and model mice were feed with the high-lipid for 5 weeks.The type 2 Diabetic renal lesion mice were devided into five groups,including untreated group,diformin（3.0mg·10g-1） therapy group,and Lycium seed oil groups（0.05,0.1 mL·10g-1）.The administration lasted for five weeks,and at the end of the experiment,the blood glucose,urine glucose,24h urine volume,24h urine protein excretion quantity were collected.The renal quality indices,renal glomerulus area,the pathological change and the interleukin-8（IL-8） level were observed,.Results The blood glucose,urine glucose,24h urine volume,24h urine protein excretion quantity,renal quality indices,renal glomerulus area and interleukin-8（IL-8） leve reduced significantly in Lycium Barbarum seed oil treatment compared to diabetic model group.Conclusion Lycium Barbarum seed oil can prevent kidney lesion in type 2 diabetic by reducing the blood glucose level,Urine glucose level,renal glomerulus area,24h urine volume and the 24h urine protein excretion quantity,and as the renal quality indices and the interleukin-8（IL-8） level as well.
