摘  要:目的观察骨形成蛋白.2(BMP-2)基因转染兔骨髓基质细胞(MSCs)复合藻酸钙修复兔膝关节软骨缺损的效果。方法取4周龄的新西兰兔骨髓细胞,体外培养得到MSCs。选用24只成年新西兰兔,在两侧股骨髁非负重区造成全层软骨缺损模型,A组(实验组)植入BMP-2转染的MSCs+藻酸钙,B组(对照组Ⅰ)植入空质粒转染的MSCs+藻酸钙,C组(对照组Ⅱ)植入不含MSCs的藻酸钙,D组(对照组Ⅲ)旷置。12周后,观察软骨缺损修复情况及组织学评价。结果实验组修复组织基本光滑,有大量Ⅱ型胶原蛋白表达,3个对照组修复组织Ⅱ型胶原蛋白表达量少,仍有明显缺损。实验组和3个对照组组织学评分差异有统计学意义(P〈0.05)。结论BMP-2转染兔MSCs修复软骨缺损效果优于其他对照组。Objective Rabbit-bone marrow stromal cells (MSCs) transfected by bone morphoge- netic protein-2 (BMP-2) gene mixed with calciumalginate to repair defects of articular cartilage of rabbit knee, then to observe the result of repairing. Methods Extract bone marrow cells from a four-week-old New Zealand rabbit. Isolated and purified bone marrow stromal cells in vitro. Choose 24 grown-up New Zealand rabbits and divide them into 4 groups randomly. Full-thickness articular cartilage defects were made in the non-weight-bearing area of the bilateral femoral condyles in New Zealand rabbits. Transfer the different four groups complex component to the different defect place of the articular cartilage : In group A, MSCs + calciumalginate complex, which has been transfected by BMP-2 (Experimental Group) ; In group B, MSCs + calciumalginate complex, which transfected by Empty plasmid (Control GroupI) ; In group C, calci- umalginate without cells (Control GrouplI) ; In group D, without any treatment (Control Grouplll). The fem- oral condyles were harvested at the end of 12 weeks, and then observe the condition of repairation of cartilage defects and make assessments of histology. Results Repair Tissue part in experimental group was almost flat smoothly, consisting of a large of type-ll collagen. In 3 control groups, the type-]] collagen of Repair Tissue was bit and had apparent defection. There were significant difference between the experimental group and the three control groups in histological evaluation (P 〈 0. 05). Conclusion The group which was transfered with BMP-2 + MSCs is superior to control groups in the effect of repairing cartilage defects.
