通心络对糖尿病小猪肾组织中TGF-β_1表达的影响

摘　　要：目的研究转化生长因子β1(TGF-β1)在糖尿病小猪肾组织中的表达及中药通心络对其表达的影响。方法以联合注射链尿佐菌素(STZ)与四氧嘧啶的方式复制糖尿病小猪模型,将动物随机分为糖尿病组(DM),通心络治疗组(TXL)和正常对照组(C)。6个月后处死动物,测量肾重/体重比值,应用免疫组化技术和western blotting技术观察肾组织中TGF-β1的表达。结果与对照组相比,糖尿病组小猪肾重/体重比值和TGF-β1的表达明显增加,通心络可显著降低肾重/体重比值和TGF-β1的表达。结论通心络通过影响糖尿病小猪肾组织中TGF-β1的表达改善糖尿病肾组织纤维化。Objective To investigate the effects of a Chinese traditional medicine, tongxinluo, on the expression of transforming growth factor-β1 （TGF-β1） in kidneys of pigs with diabetic nephropathy. Methods The diabetic pig models were established by injection of streptozotocin （STZ） and alloxan. The pigs were randomly divided into diabetic group （DM） , Tongxinluo-treated group （TXL） and control group （C） . The ratio of kidney weight to body weight in each pig was measured after six months. The expres- sion of TGF-β1 was detected by immunohistochemical stainning and Western blotting. Results The ratio of kidney weight to body weight and the expressions of TGF-β1, were increased compared with those in the control group, but Tongxinluo reduced the increasing. Conclusions The Chinese traditional medicine, Tongxinluo, can reduce the fibrosis in pigs with diabetic nephropathy, and its mechanism is probably related to the down-regulated expression of TGF-β1
